Phryma leptostachya L.is also known as medicinal herb,fly poisonous weed,and is a perennial herb belonging to the genus Phryma leptostachya.The medicine part of P.leptostachya L.is the whole plant has a wide range of sources,strong adaptability,rapid growth and a large planting area in China.P.leptostachya L.has the effect of clearing heat and detoxifying,its bitter taste can treat sores,the roots of the osseospermum can kill fly maggots,the whole grass can kill vegetables and vegetable worms,both seeds and stems can be used as medicines,there are blood circulation,Diuretic and detoxifying effect.Fresh grass can be smashed and applied externally,which can cure sores,swelling,pain and poisonous insect bites.After decoction of Bone Perennial Grass,it has anti-inflammatory and bactericidal effects for external use,and is suitable for general sore ringworm.Bone grass can also treat rheumatic pain,the onset of swelling and poison,rheumatoid arthritis and other symptoms.In this paper,the chemical constituents and biological activities of the whole plant of P.leptostachya L.by Shandong Dongying Company are studied,which will help to make full use of the P.leptostachya L.resources and explore its potential medicinal value.In this study,we use the whole plant of Phryma leptostachya L.by Shandong Dongying Company as the raw material.Column chromatography and HPLC methods were used for separation and purification.The structures of the compounds were identified by NMR spectroscopy data.Tricin(1),tricin 4′-O-(threo-β-guaiacylglyceryl)ether(2),tricin 4′-O-(erythro-β-guaiacylglyceryl)ether(3),(+)-isolariciresinol(4),sesquipinsapol B(5),methyl caffeate(6),4-[(3,4-dihydroxycinnamoyl)-oxy]-methyl cinnamate(7),phthalic acid bis-(2-ethylhexyl)ester(8),5-(methoxymethyl)-1-(4-hydroxyphenethyl)-1H-pyrrole-2-carbaldehyde(9).Nine compounds were isolated and identified,of which 1-6,9 were the first known compounds in the genus.In the DPPH system,compounds 3-6,8-9 have stronger free radical scavenging activity,and compounds 1,2,7 have weaker activity.In the ABTS system,the compounds with strong free radical scavenging activity are compounds 4 and 6,and the compounds with weaker activity are 1-3,5,7-9.The isolated compounds 1-9 were induced by H2O2 using MTT method.Study on the neuroprotective activity of SH-SY5 Y cell injury.Compounds 1-9 all exhibit potential neuroprotective activity.Among them,the cell survival rates of compounds 2,6 and 7 were significantly enhanced of 81.68%(50 μM),92.15%(50 μM)and 93.19%(50 μM),respectively,comparing with the cell viability of the positive control drug vitamin E(76.55%,50 μM).Studies have shown that compounds 1-9 may be potential drugs for the treatment of neurodegenerative diseases.Compounds 6,7,and 9 exhibited strong Ach E with IC50 of 34.43,39.61,and 27.48μg/m L,respectively.Compared with the positive control galantamine hydrobromide IC50 was 23μg/mL.
