The lipid peroxidation reaction caused by the active oxygen produced by the body and PUFA in biological membrane phospholipids.Studies have shown that lipid peroxidation have closely related to pathological process,such as tumor,chemical poisoning,infection,inflammation,autoimmune disease,Parkinson disease,atherosclerosis(AS),cardiovascular and cerebrovascular diseases and causality.Body can resist lipid peroxidation damage through intaking of food and drugs which containing anti-lipid perxidation active substances.we use two-way fermentation of the monascus and Dendrobium nobile Lindl in order to produced active material has stronger ability to anti-lipid peroxidation in the interaction,for the development of new anti-lipid peroxidation product of two-way fermentation to provide a certain basis.The study adopts two-way fermentation of the monascus and Dendrobium nobile Lindl,It used three antioxidative mode in vitro of scavenging ABTS radical,scavenging DPPH radical and Inhibiting lipid peroxidation to screened strain has the high oxidation activity from two-way fermentation of Monascus and Dendrobium nobile Lindl.It analysis secondary metabolites' contents of alkaloids,polysaccharides,polyphenols,determine the change of contents in secondary metabolites in medicinal fungus qualitative.Using biological activity tracking method,through the analysis activity of anti-lipid peroxidation in vitro,to study the extraction of anti-lipid peroxidation component and the distribution of the active component;Using column chromatography to separate coarse extraction products to get different components,by the analysis activity of anti-lipid peroxidation in vitro to determine active components.And by using silica gel column chromatography to separate and purificate component to get the monomer has anti-lipid peroxidation activity.The main results as follows: secondary metabolites in medicinal fungus qualitative.Using biological activity tracking method,through the analysis activity of anti-lipid peroxidation in vitro,to study the extraction of anti-lipid peroxidation component and the distribution of the active component;Using column chromatography to separate coarse extraction products to get different components,by the analysis activity of anti-lipid peroxidation in vitro to determine active components.And by using silica gel column chromatography to separate and purificate component to get the monomer has anti-lipid peroxidation activity.The main results as follows: 1.It used three antioxidative mode in vitro of scavenging ABTS radical,scavenging DPPH radical and Inhibiting lipid peroxidation to screening strain which two-way fermentation has higher activity of anti-lipid peroxidation.The polysaccharide: The content of Polysaccharides in bi-directional fermentation products,red yeast rice and Dendrobium nobile Lindl were analyzed by phenol sulfuric acid method,results indicated that the content of Polysaccharides in bi-directional fermentation is higher than red yeast rice,the contents of polysaccharides are 3.15,2.1,1.73mg/g respectively.The polyphenol:The content of polyphenols in bi-directional fermentation products and red yeast rice were analyzed by Folin-Ciocalteus method,Results indicated that the content of polyphenols in bi-directional fermentation is higher than red yeast rice,the contents of polyphenols are 2.03,1.5 and 0.08mg/g respectively,with a significant difference(P<0.01).The alkaloid: the contents of alkaloid are 2.79 and 1.79mg/g respectively,with a significant difference(P<0.01).2.Through the activity analysis of anti-lipid peroxidation in vitro,using different concentrations of ethanol to extract anti-lipid peroxidation component in medicinal fungus,determine the water and 90% ethanol solution as the best extraction solvent to extract anti-lipid peroxidation component in medicinal fungus.The method of active tracking shows that the anti-lipid peroxidation component in medicinal fungus distribute in ethyl acetate parts in the extracts of alcohol crude products.3.Let extracts of water and alcohol crude products in the medicinal fungus through silica gel column chromatography and neutral alumina column chromatography respectively for separation and purification,get six components,the active part tracking method shows that anti-lipid peroxidation effect of component 4 is strongest,followed by component two.You can see component 2 from the thin-layer chromatography,relative to other components,component 2 is easy to isolate single active substances.Adopting the silica gel column chromatography to further separation and purification to get active monomers.4.Using mass spectrometry(mass spectra and MS),nuclear magnetic resonance(nuclear magnetic resonance,NMR)spectrum technique to identify compounds,results show that the monomer is X.
