Dielectrophoresis is a method to detect the activity of biological particles on a dielectrophoresis chip.The test uses separate devices currently,and the combination between instruments is poor,which cannot exploit the advantages to the full and be easy to develop.At the same time,manual evaluation of electrophoretic video is labor-intensive and unreliable,which cannot meet the automation requirements.To address the above problems,an automatic device which integrates the functions of bio-chip,special power supply,micro-video system and PC is designed.It uses digital image processing algorithm to realize the automatic detection of video images.The main research works are as follows:First of all,in order to solve the problem of poor coordination between instruments,a new scheme is proposed to break the existing structure for integrating the functions of the instruments.Based on the original work,the paper is mainly designed for the control system.The basic peripheral circuit,interfaces,drivers and application software are designed to realize the basic function and hardware of the system.Secondly,in order to solve the problem of manual evaluation,a scheme is proposed using image processing instead of the human eye.The image processing algorithms,which targets the most common test object – cell,are divided into static processing algorithms and dynamic processing algorithms to realize the automatic detection of cell speed and trajectory.Static processing algorithms consist of five steps: binarization,morphological filtering,object detection,centroid calculation and feature point marking.The dynamic processing algorithms consist of three parts: cell tracking,trajectory description and speed calculation.The SURF algorithm is optimized for feature point tracking detection.A new method combined the Harris corner detection and L-K optical flow method for feature point tracking detection is proposed.It requires better hardware,detection accuracy is also better.Finally,combined with the system application requirements,tests are designed.The cell video image detection test was carried out by Connected of the control system,the power module and the micro-video module.The relative error between the motion parameter detection and the actual parameters was within 1%.In summary,the dielectrophoresis integrated system's structural integrated design and the design of the host control system based on the Android platform have been completed.They simplify the complex biochip experiment,and realize miniaturization and integration of the system,which provides a possibility to build the Internet of laboratory instruments in the future.
