Objective: This work studied the lipid droplets(LDs)of one intestinal actinomycetes,Streptomyces thermovulgaris(S.thermovulgaris),by analyzing the peripheral proteins on LDs of S.thermovulgaris,and extending to discover the distribution of intestinal actinides owning LDs in human intestinal system.Furthermore,by analyzing the variation in absolute abundance of intestinal bacteria which might contain LDs in athlete's feces under different exercise intensities,we were able to provide valuable clues to the study of relationship between intestinal bacteria and exercise as well as health clue.Methods: The intestinal actinomycetes S.thermovulgaris were cultured in vitro using microbiological methods.LDs were found in S.thermovulgaris using quantitative lipid analysis and cell morphology.Then,using biophysical and biochemical method like density gradient centrifugation,the LDs of S.thermovulgaris were isolated are purified.SDS-acrylamide gel electrophoresis,and silver staining of proteins were applied to analyze the LD proteome of the bacteria,confirming the existence of LDs.Finally,the bioinformatics methods were utilized to discover more bacteria owning LDs in the intestinal system,and further to analyze the variations in the abundance of actinomycetes in athlete's feces under different exercise modes and intensities.Results:(1)The S.thermovulgaris in the intestinal tract contains triglycerides.(2)After the staining of intestinal S.thermovulgaris using neutral lipid stain,the lipid droplet-like structures can be observed inside the bacteria using microscopy.(3)Multiple proteins reside on the surface of LDs from intestinal actinomyces S.thermovulgaris.(4)Elongation factor thermo unstable(EF Tu)is a LD protein of S.thermovulgaris,and its homologous proteins exist in multiple intestinal actinomycetes.(5)The absolute abundance of two Actinomyces bacteria collected from the feces of swimming athletes shows regular changes when the athletes undertake different exercise intensities.Conclusions:(1)The intestinal bacteria S.thermovulgaris contain lipid droplets.(2)All 87 actinomycetes collected from human gut may contain LDs.(3)During long-term sports training,the absolute abundance of two Actinomyces bacteria that may contain LDs in the feces of athletes increase firstly and then decrease.
