| Objective: This paper attempts to explore the effect of aerobic exercise on 5-HT in the medial prefrontal cortex of Parkinson's Disease rats,and to explore the mechanism of the neuroprotective effect of 5-HT induced by aerobic exercise on PD rats.It reveals that aerobic exercise is a beneficial health treatment scheme and is expected to provide reference for the research and prevention of PD and other degenerative diseases.Methods: 48 3-month-old SPF grade healthy male SD rats were randomly divided into four groups: SEDS group and ex normal saline exercise group;sed-pd Parkinson's quiet group and ex-pd Parkinson's exercise group.On the basis of Bedford's modeling scheme and previous research of the research group,the following exercise scheme was designed for the rats.A 3-day adaptive treadmill training was conducted in advance(the speed was 10 meters per minute,the first day's exercise time was 10 points,the second day's exercise time was 20 points,the third day's exercise time was 25 points,and the gradient was 0 °).After one day's rest,the rats were ready for formal training the next day.The feeding box is made of plastic material,stainless steel cover,plastic water bottle and stainless steel metal water pipe.The rats are fed according to the national standard sentinel animal feed.The temperature is maintained at 20℃~ 25℃ and the relative humidity is 45% to 55%.Eight weeks of cardio.At the end of eight weeks of aerobic training,one week of modeling was carried out,and 6-OHDA was injected into the right side of the striatum of the rat brain.After two weeks of rehabilitation exercise,as well as rotation experiment detection and evaluation,materials were taken 24 hours after the last rotation experiment,and the morphological changes of medial prefrontal lobe were observed by HE staining.The contents of 5-HT and 5-HT1 A,the activities of TPH2 and MAOA were detected by ELISA.Immunohistochemistry was used to detect TH and A-synuclein protein expressions.Results: the success rate of pd-sed group and pd-ex group was 77.78% and 71.43%,respectively.He staining: the number of neurons in sed group was more,arranged orderly,and the morphology of cells was normal.However,in sed-pd group,the number of neurons in medial prefrontal lobe was relatively small,and the arrangement was disordered;The results of ELISA showed that the contents of 5-HT and 5-HT1 A in the prefrontal cortex of sed-pd group were significantly lower than that of SED group(P < 0.01);the activity of TPH2 in the prefrontal cortex of sed-pd group was significantly lower than that of SED group(P < 0.01);the contents of 5-HT and 5-HT1 A in the prefrontal cortex of ex-pd group were significantly higher than that of sed-pd group(P < 0.01).Compared with sed-pd group,TPH2 activity in prefrontal cortex of ex-pd group was significantly higher(P<0.01).IHC results showed that the expression of th in the striatum of sed-pd group was higher than that of SED group.Compared with sed and sed-pd,the expression of α-synuclein in ex group and ex-pd group decreased significantly.Conclusion: aerobic exercise can up regulate the 5-HT content in medial prefrontal lobe of Parkinson's rats;it may be that exercise can activate the activities of TPH2 and MAOA;increase the synthesis of 5-HT,reduce the metabolism of 5-HT,enhance the expression of 5-HT1 A,and improve the function of 5-HT nervous system. |
