Objective:In this experiment,a nutrition-obese rat model and a hypoxia endurance exercise model were established.Three kinds of hypoxic concentrations(11.3%,13.3,and 16.3% oxygen concentration)combined with endurance exercise were used to intervene in obese rats and passed Western Blot.Real-time fluorescence quantitative PCR was used to detect m RNA expression of autophagy regulators Sestrin2,AMPK?2,and autophagy factors Beclin1 and LC3 II.The effect of hypoxia or endurance exercise on autophagy-related factors in skeletal muscle cells was explored to explore the role of hypoxia exercise in weight loss.Control or reduce autophagy-related diseases provide experimental evidence.Methods:The model of nutritional obesity rats fed with high fat diet for 8 weeks was established and randomly divided into 8 groups: normoxia group(A),normoxia group(AE),16.3% hypoxia(BE),BE(13.3%),CE(13.3%),Hypoxia Quiet(11.3%),Hypoxia Group(DE),each group of 10,and continue to feed high-fat diet.Formal experimental intervention period,the rat training program is as follows: exercise group rats were moderate-intensity endurance exercise,wherein the time is 40 min,running table slope is 00,running speed of 20 m / min;exercise frequency of 5 times a week Friday)for 8 weeks.After the last training,fasting for 24 hours,rats were sacrificed and sampled.Biochemical indicators of blood lipid and blood glucose concentration;Western Blot measured skeletal muscle cell autophagy related proteins Sestrin2,AMPK ? 2,Beclin1 expression levels and LC3 II / LC3 I ratio;real-time fluorescent quantitative PCR assay autophagy regulatory factors Sestrin2,AMPK ? 2and The expression of Beclin1 and LC3 II m RNA were observed.The changes of morphology and structure of skeletal muscle cells were observed by paraffin sections and HE staining.Results:1.The rat model of nutritional obesity: After 8 weeks of high-fat diet,the body weight,Lee's index,BG,TC,TG and LDL-c concentrations were significantly increased(P <0.05).2.Compared with group A,the body weight,BG,TC,TG and LDL-c concentrations in AE group were significantly decreased(P <0.05)and HDL-c concentrations were significantly increased(P <0.05).The concentrations of TG and BD in LDL-c,B,C and D group were significantly decreased(P <0.05)in group D,HDL-c concentrations in group C and D were significantly increased Compared with AE group,the concentrations of LDL-c and TG in DE group were significantly decreased(P <0.05),the concentrations of BG in CE group and DE group were significantly decreased(P <0.05),while the concentrations of HDL-c in DE group were significantly increased <0.05).3.Western Blot results showed that compared with group A,the protein expression of Sestrin2,AMPK?2,Beclin1 and the ratio of LC3 II / LC3 I were significantly increased in AE group(P <0.05);In group C and D,Sestrin2,(P <0.05).The protein expression of Beclin1 in group B,group C and group D was significantly higher than that in group AE(P <0.05).Compared with group AE,CE and DE The protein expression of Sestrin2 and AMPK?2 and the ratio of LC3 II / LC3 I were significantly increased(P <0.05).The protein expression of Beclin1 in DE group was significantly increased(P <0.05).4.Real-time quantitative PCR results showed that the m RNA expressions of Sestrin2,Beclin1 and LC3 II in skeletal muscle cells of AE group,C group and D group were significantly higher than those of A group(P <0.05)(P <0.05).Compared with AE group,m RNA expression of Beclin1,LC3 II and AMPK?2 in CE group and DE group were significantly increased(P <0.05),and m RNA expression of Sestrin2 in DE group was significantly increased P <0.05).Conclusion : 1.Three different concentrations of hypoxia or combined endurance exercise can reduce body weight in obese rats and improve blood glucose,lipids and skeletal muscle cell morphology,structure,hypoxia and exercise together to reduce the body weight of rats,blood glucose,blood lipids And skeletal muscle cell morphology and structure improvement effect is more significant.2.Endurance exercise,hypoxia exposure,and hypoxia combined with endurance exercise can induce autophagy in skeletal muscle cells;and the cumulative stimulation effects of exercise and hypoxia are more prominent than simple endurance exercise and hypoxia exposure.3.Endurance exercise,hypoxia exposure,hypoxia combined with endurance exercise can increase the expression of Sestrin2,AMPK ? 2,Beclin1,LC3 II factor,improve LC3 II / LC3 I ratio,thus effectively activating and enhancing autophagy in skeletal muscle cells of obese rats,especially Is 11.3% Hypoxic tolerance exercise group more obvious effect.
