1 ObjectiveIn this study,heart-specific silencing of Ca-?1D gene in Drosophila melanogaster was performed using UAS-Gal4 system.Drosophila hypoxia and exercise intervention were performed using Drosophila hypoxia device and exercise device.To investigate the effect of hypoxia on the cardiac function of heart-specific silencing Ca-?1D gene in Drosophila melanogaster.Meanwhile,exercise capacity,sleep status and life cycle were measured.In order to study the mechanism of hypoxic exercise intervention on cardioprotection,and to provide a scientific basis for the connection between L-type calcium channels in cardiomyocytes.2 Materials and methodsThe virgin flies with genotype w1118 and P {GD1737} v51491 were crossed with W1118 and Hand-GAL4 male flies to obtain FI generation of virgin flies,and were divided into 8 groups:normoxemia group(NC)HC,NE,Hypoxic exercise group,NCS,HCS,NES,Hypoxic exercise disturbance group(HES).Using the third generation of Drosophila motion device,exercise group were 22 days of age began to exercise,each training 1.5h,continuous training for two weeks.Hypoxia group began hypoxia on the 22nd day of age,hypoxia each time from 7:00 pm to 7:00 the next morning,hypoxia 12h,eontinuous hypoxia for two weeks.Index Detection:60 Drosophila were randomly selected from the two constructed strains for RT-PCR test.Each group was randomly selected 30 Drosophila M-mold heart function test,32 Drosophila sleep monitoring,100 Drosophila climbing ability test,200 Drosophila statistical life cycle.3 Results3.1 The result of RT-PCRCompared with the W1118/w1118;P {GD1737} v51491 group,the expression of Ca-?1D gene mRNA decreased(P<0.01)in Hand-GAL4/wlll8 andP {GD1737} v51491 group,and the over-expression rate was-48.15%.Described FI generation of Drosophila Ca-?1D gene silencing line was constructed successfully.3.2 The result of M-mold cardiac functions in drosophilaCompared with the NC group,the shortening fraction of the fruit fly in the NCS group decreased(P<0.05),fibrillation had no significant difference(P = 0.81),and the heart rate index increased(P<0.01).Tip Ca-?1D gene downregulation of heart blood pump ability to reduce the risk of irregular heartbeat.Compared with NC group,the shortened fraction of HC group,NE group and HE group increased(P<0.05),fibrillation of HE group decreased(P<0.05).The fibrillation in NE group was higher than that in HE group(P<0.05),and the heart rate index was higher than HC group and HE group(P<0.05).Compared with NCS group,the shortened scores of HCS group,NES group and HES group increased(P<0.01),the fibrillation of NES group and HES group decreased(P<0.05),HES group heart rate index decreased(P<0.01).Compared with NES group,the index of heart rate irregularity in HCS group decreased(P<0.05),while the index of heart rate irregularity in HES group decreased(P<0.01).3.3 sleep monitoring resultsCompared with the NC group,NCS night sleep reduced(P<0.01),night activities increased(P<0.01),daytime sleep no significant difference(P>0.05),daytime activities decreased),No significant difference in total sleep time(P>0.05),the average number of activities per hour Was no significant difference(P>0.05).Compared with NC group,nighttime sleep time of HE group increased(P<0.01),night activities of HC group,NE group and HE group decreased(P<0.05),but there was no significant difference between daytime sleep group and HC group The activity of oxygen in exercise group decreased day by day(P<0.05),and the total sleep time increased in HE group(P<0.05).The average number of activities per hour decreased in HC group,NE group and HE group(P<0.01).Compared with NCS,HES group increased night sleep time(P<0.01),HCS group,NES group and HES group decreased night activity(P<0.01),daytime sleep time and activity frequency had no significant difference in each group,HES group Total sleep time increased(P<0.01),and the total sleep time in HES group was higher than that in interference group,HCS group and NES group(P<0.01),and the average hourly activities in NES group and HES group decreased(P<0.01).The average number of activities per hour in HES group was lower than that in HCS group(P<0.05).3.4 climbing ability monitoring resultsThe climbing ability of NCS group was significantly lower than NC group(P<0.01).Compared with NC group,the climbing index of HC group increased significantly(P<0.01),and HC group was significantly higher than that of NE group and HE group(P<0.05).Compared with NCS,the climbing index of HCS group was significantly higher(P<0.05),and HCS group was significantly higher than that of HES group(P<0.05).3.5 Results of the lifespan statisticsCompared with NC group,life span of NCS group was significantly shorter(P<0.05).Compared with NC group,HC group had longer life span(P<0.05)and longer life span(P<0.01).And the life span of HE group was longer than that of NE group(P<0.01).The life span of NES group was longer than that of NCS group(P<0.05),and the life span of HCS group and HES group was significantly longer than that of NCS group(P<0.01).4 Conclusion4.1 The heart-specific Ca-?1D gene silencing of Drosophila strain was successfully constructed by UAS-Ga14 system.It was found that cardiac-specific Ca-?1D gene silencing can lead to impaired cardiac pump function and heart rate in Drosophila.Fatigue life reduced,and will lead to decreased exercise capacity and quality of sleep.4.2 Under different genetic backgrounds,Ca-?1D gene expression down-regulated Drosophila and normal expression of Drosophila after hypoxia intervention in the heart pump function and heart rate irregularities have been improved,sleep quality is improved,and exercise capacity,life expectancy Extend has a positive effect.4.3 Hypoxic exercise intervention for heart function,sleep quality and longevity of Drosophila better than hypoxia or exercise a single intervention,but did not increase Drosophila athletic ability.Hypoxic exercise intervention can not only improve the age-related decline of normal expression group,but also improve the heart-specific Ca-?1D gene expression down-regulation of heart function damage and quality of life of fruit flies.
