Purpose:In order to provide sicentific theroy basis of inducing the muscle injury in HiLo, we investigate the effect of hypoxia on myocardial membrane injury after the eccentric exercise.Methods:20 male SD rats,8 years old, undertooka high load and interval running. The training is the eccentric exercise of 26.8m/min and 5min X 10 groups, which rests lmin between groups. After that, these rats rested in the 12.7% oxygen concentration environment. And then randomly dividedinto two groups:normal saline group (C group) and Calpain inhibitor group (I group). To exa.med cell membrane permeability, two rats were chosen randomly to inject EBD. What’s more, the rest of rats were injected normal saline and Calpatatin inhibitor, and then we measured the protein levels of Calpainl large subunit, Calpainl small subunit and Calpain2 by western blot.Results:(1) The positive cells rates of rats gastrocnemius in I group is lower than C group(P<0.01). (2)The protein levels of Calpainl 80kDa large subunit, Calpainl 30kDa small subunit and Calpain2 in I group are remarkebly lower than C group(P<0.01).Conclusions:Injecting the inhibitor after one-off eccentric exercisecan suppress Calpainl&2 expression in hypoxia, as well as slowing down the cell membrane pentrability. It proves that Calpainl&2 are playing important roles in hypoxic mechanism of myocardial membrane.
