Observation Of Curative Effect Of MEBT/MEBO On Wounds After Anal Fistula And The Effect Of BMP-7 Expression

Abstract/Summary:
Objective: To observe the clinical effect of MEBT / MEBO on wound healing after anal fistula and the expression of BMP-7 in rat wound tissues.Methods:(1)90 patients who undergoing surgery in our hospital from May 2018 to September 2019 were randomly divided into 30 cases in the control group,30 cases in the MEBO group,30 cases in the rb-b FGF group.The MEBO group was treated with MEBT/MEBO,and the rb-b FGF group was treated with Recombinant Bovine Basic Fibroblast Growth Factor,the control group was treated with Saline.The secretion,granulation tissue and anal pain on the 3rd,7th,14 th and 21 st days after operation of the three groups were compared.The wound healing of the three groups on the 2nd,14 th and 21 st days after operation was compared.(2)54 healthy adult SPF SD male rats were randomly divided into the Model group(18 rats),the MEBO group(18 rats)and the rb-b FGF group(18 rats)according to the random number table method.After the model was established,the control group was treated with saline,MEBO group were treated with MEBO,and rb-b FGF group were treated with Recombinant Bovine Basic Fibroblast Growth Factor.Wound tissues were taken for detection on days 3,7,and 14 after treatment.Western blot was used to detect the expression level of BMP-7 protein in the wound tissue of each group.Oil red O staining was used to observe the formation of adipocytes in the wound tissue.Hematoxylin-eosin staining was used to observe the wound healing.Masson stain Observe the fibrosis of the wound tissue.Results: 1.There was no significant difference in wound secretions between the three groups on the 3rd day after operation(P> 0.05),and the least on the 7th day after the operation in the rb-b FGF group,on the 14 th and 21 st days after the operation The wound secretions of the group and the rb-b FGF group were less than the control group(P <0.05);2.On the 7th,14 th,and 21 st days after surgery,the granulation tissues of the MEBO group and the rb-b FGF group were superior to the control group;3.There was no significant difference in the scores of anal local pain between the three groups on the 3rd postoperative day(P> 0.05).On the 7th,14 th,and 21 st postoperative day,the pain level of the experimental group and the rb-b FGF group was lower than that of the control group(P <0.05);4 The wound healing rate of the experimental group and rb-b FGF group was higher than that of the control group on the 14 th and 21 st days after operation(P <0.05).Conclusion: The skin regenerative medical technology can effectively reduce the pain of the operation port,reduce the exudation,and accelerate the healing of the wound after anal fistula.(5)On the 3rd,7th,and 14 th day of treatment,the BMP-7 expression level in the wound tissue was statistically significant,and the BMP-7 expression level was gradually increased(Day 3 <Day 7)<Day 14),Comparison between groups: there were statistically significant differences between MEBO group,rb-b FGF group and Model group(P<0.05),while there were no statistically significant differences between MEBO group and rb-b FGF group(P>0.05).(6).With the prolongation of treatment time,the number of adipocytes increased gradually(P<0.05),and the number of adipocytes in MEBO group and rb-b FGF group was more than that in Model group(P<0.05).Conclusion:(1)MEBT/MEBO can effectively reduce the pain of the operation port,reduce the exudation,and accelerate the healing of the wound after anal fistula.(2)MEBT/MEBO can effectively affect the expression level of BMP-7 in rat wound tissues and promote the formation of adipocytes in wound tissues,and inhibit excessive fibrosis of wound tissues.
