Correlation Of Polymorphisms In MFG-E8 Gene With Coronary Artery Disease In Han Population In North Guangxi,China

Objective: To investigate whether the MFG-E8(milk fat globule EGF factor?)gene single nucleotide polymorphism is related to coronary artery disease.Methods: A case-control study was used to select 158 patients with coronary artery disease and 183 healthy controls.Two polymorphisms in MFG-E8(rs 4945C>A and rs 1878326 C>A)were analyzed by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP)and DNA sequencing in 158 Coronary artery disease patients and in 183 controls.The serum MFG-E8 levels in the coronary artery disease group and the control group were detected by ELISA.The relationship between serum MFG-E8 levels and coronary artery disease was analyzed,and the correlation between serum MFG-E8 concentration and genotype in patients with coronary artery disease was analyzed.Results: The serum concentration of MFG-E8 in the coronary artery disease group was significantly lower than that in the control group(Z=-15.370,P<0.001);The polymorphism of MFG-E8 gene rs 4945 C>A locus is associated with coronary artery disease.(CA genotype vs CC genotype: OR=2.029,95%CI: 1.229-3.349,P=0.006;A allele vs C allele: OR=1.835,95%CI: 1.153-2.921,P=0.010);No correlation found between rs 1878326 and coronary artery disease;The rs4945C>A genotype did not affect MFG-E8 serum concentration(Z=-1.580,P=0.114).Conclusions: 1.Negative correlation between serum MFG-E8 levels and coronary artery disease in the Han population in North Guangxi.2.Among Han people in northern Guizhou,MFG-E8 gene rs 4945 C>A locus polymorphism may be related to coronary artery disease.3.Among the Han people in North Guangxi,the MFG-E8 gene rs 1878326 C>A locus polymorphism is not related to coronary artery disease.4.No correlation was found between MFG-E8 gene rs 4945 C>A polymorphism and serum MFG-E8 level in the coronary artery disease group of Han people in North Guangxi.
