Obesity is a metabolic syndrome that causes many chronic diseases worldwide,leading to reduced life expectancy,cardiovascular disease,type 2 diabetes,non-alcoholic fatty liver disease(NAFLD),and some other type of complications.Dietary fiber is a food ingredient associated with reduced appetite and weight.Sufficient dietary fiber in the diet can prevent excessive food intake and fat accumulation.Recently,we have found a kind of polysaccharide-flaxseed polysaccharide(FP)with anti-obesity effect.It is rich in dietary fiber and hasbeen reported of many functional properties.But the anti-obesity mechanism and the bioaccessibility of FP remains to be elucidated.Firstly,the signal pathway of linseed polysaccharide against obesity in vivo was analyzed;then,by comparing the gene expression differences between obese rats and FP intake rats,we explored the effect of FP on obese rats in intestinal tract based on transcriptome,screened out other differential genes related to energy metabolism and verified them by q PCR.The results are as follows:1)After FP intervention,the body weight and abdominal fat coefficient of obese rats were significantly reduced from 530±16 g and 2.15%±0.30%(HFD group)to 478 ± 10% G and 1.38%±0.48%(FPD group);intake of FP reduced the levels of TNF?,IL6 and IL? in serum of obese rats(P < 0.05),and alleviated the level of liver inflammation;the NPY protein expression decreased significantly(P < 0.05)and the GLP-1 protein expression increased significantly(P < 0.05),which made the appetite of obese rats return to normal;while the increase of adiponectin stimulated the expression of AMPK,down regulated the expression of ACC,SREBP-1 and FAS,up regulated the expression of AMPK,HST,ATGL and CPT-1.2)The intervention of FP increased the expression of 14 genes related to energy metabolism,such as Car3,Cox4i2 and Ndfa4l2.Conclusion: FP upregulates the energy metabolism related genes such as car3 in intestinal tissue,and promotes energy consumption.Among them,up-regulation of Cox4i2 and Ndufa4l2 protein expression and up-regulation of adiponectin may promote AMPK signaling pathway to improve lipid metabolism,including improving lipolysis and fatty acid oxidation and inhibiting lipogenesis.The upregulation of AMPK signaling pathway,together with the elimination of leptin resistance by inhibiting the production of inflammatory factors and the restoration of satiety through the significant downregulation of NPY and the upregulation of GLP-1,reduced the weight of rats to a normal level.
