Research Of The Clinical Effect Of Tangyipingfang In Treating Patients With IGR And Its Intervention On The AMPK Signaling Pathway Of Wistar Rat With IGR

Objective: To observe and analyze the clinical effect of Tangyipingfang which takessoothing liver and promoting blood flow and eliminating phlegm as the rules of treatmenton patients with IGR and discuss its influence on the AMPK signaling pathway throughexperiments.Methods:60patients with clinical diagnosis of IGR were randomly divided intoTangyiping group (n=30) and the control group (n=30). Besides receiving the same normaltreatment, Tangyiping group received Tangyipingfang treatment on its base, while thecontrol group received melbine treatment on the same base. In the three-month observation,the change of the following indexes were closely recorded as body weight index, WHR,the clinical symptoms, FPG and PBG, insulin, IRI, ISI etc. Experiments were conducted tocompare the data of the following indexes as the general condition, body weight, bloodglucose, insulin, IRI, ISI and the expression of AMPKα, SAD-A protein and mRNA in thepancreatic tissue cells of the wistar rat in the normal wistar rat group, IGR model wistar ratgroup, TCM wistar rat group and the melbine wistar rat group.Results: Clinical results suggested that Tangyiping group P＜0.01in BMI, WHR, FPG andPBG, insulin,and P＜0.05in IRI, ISI and scores of TCM symptom compared with those ofbefore the treatment. In the evaluation of the curative effect, the TCM Tangyipingfang P＜0.01in the IGR curative effect, the efficacy of TCM symptoms and the comprehensiveefficacy compared with those of before the treatment and P＜0.05compared with melbine.And there was no adverse reaction during the tests. Experiment results suggested that Tangyiping wistar rat group P＜0.05in the blood glucose and ISI，P＜0.01in insulin andIRI compared with Model group. compared with the model group, the proteinquantification of AMPKα and expression of mRNA in the pancreatic tissue cells P＜0.05;compared with the model group, the protein quantification of SAD-A and expression ofmRNA in the pancreatic tissue cells P＜0.01.Conclusion: Tangyipingfang has a significant curative effect on the patients with IGR. Itcould obviously reduce the body weight index, WHR and blood glucose of the patientswhile improve the insulin resistance and could significantly improve the patients’ clinicalsymptoms. The experiment has proved Tangyipingfang could improve the insulinresistance by influencing the AMPK signaling pathways. The drug can be safely usedwhich proved that applying TCM dialectical therapy to treat IGR will have a broadprospect.
