Ovarian cancer(OC)is a common tumor in the female reproductive system,with a high mortality rate,ranking first in gynecological malignancies.Related studies have found that histone modifications in epigenetics are closely related to the occurrence and development of tumors,and the expression of ASH2 L,an important component of the core subunit of the MLL methyltransferase complex,is closely related to the proliferation and metastasis of malignant tumors However,the correlation between ASH2 L and ovarian malignancies has not been confirmed by related studies.This study explores the expression changes of ASH2 L in ovarian tissues of epithelial ovarian cancer,aiming to provide a valuable research basis for its clinical value and immune function research.Objective Observe the expression of ASH2 L,an important component of the core subunit of the MLL methyltransferase complex,in ovarian malignant tumors,and explore the effect of ASH2 L expression on the proliferation of ovarian malignant tumor cells SKOV-3,with a view to providing new diagnosis and treatment Ideas.Methods From March 2018 to July 2019,20 patients who underwent surgical treatment in the Department of Gynecology,Second Affiliated Hospital of Anhui Medical University due to ovarian cancer and benign diseases of the postmenopausal uterus during the same period were selected as the research objects.All ovarian tissues were collected and immunohistochemistry was used.The localization of ASH2 L protein expression in ovarian tissue was detected experimentally;Western blot was used to detect the expression of ASH2 L in ovarian malignant tumor tissues and normal ovarian tissue specimens,and the expression of ASH2 L in ovarian malignant tumor cell lines;mediated by polyplus reagent ASH2 L plasmid transfection to verify the transfection efficiency of ASH2 L.The CCK-8 method was used to detect the proliferation of SKOV-3 cells after transfection.Results ASH2 L protein expression is located in the nucleus of ovarian cancer tissues.Compared with normal human ovarian tissue,the expression level of ASH2 L protein in epithelial ovarian cancer tissue was up-regulated(P<0.01),and compared with 293 T cell line,the expression level of ASH2 L protein in SKOV-3 cell line was up-regulated(P<0.05).After SKOV-3 cell line wastransfected with ASH2 L plasmid,the expression of ASH2 L protein in the cells increased.Compared with SKOV-3 cells transfected with empty plasmid,SKOV-3 cells transfected with ASH2 L plasmid increased proliferation(P<0.01).Conclusions The level of ASH2 L expression in ovarian malignant tumor cells was significantly higher than that of normal ovarian cells.The level of ASH2 L gene expression can significantly promote the proliferation of ovarian malignant tumor cells.
