Effect Of Zinc Gluconate On Inflammatory Reaction In Preeclampsia-like Rats

Objective: To explore the protective role of zinc gluconate in pre-eclampsia(PE)and the potential mechanism of PE prevention,a PE-like rat model was developed by using Nitro-L-Arginine Methyl Ester(L-NAME)and was treated with zinc gluconate.Then changes of parameters reflecting the oxidative stress,inflammation,and apoptosis-related indicators,as well as the birth outcomes were measured.Methods :(1)Experimental groups: 40 pregnant rats were randomly divided into four groups,10 in each,namely the blank control(lavage and subcutaneous injection of normal saline),the treatment control(lavage of zinc gluconate 5 mg/kg/d and subcutaneous injection of normal saline),the PE model(lavage of normal salineand and subcutaneous injection of L-NAME 80 mg/kg/d),and the PE treatment(lavage of zinc gluconate and subcutaneous injection of L-NAME).(2)PE-like model indicators: blood pressure and urinary protein were measured twice in the early(including the baseline measurement),middle,and late pregnancy,respectively.(3)Birth outcome: the animals were sacrificed on the 20 th day of pregnancy.Blood and placenta were collected and the weight of the placenta and fetal rats and the length of fetal rat bodies and tails were measured.(4)Oxidative stress indicators: Antioxidant factors(CAT,GSH,SOD)and oxidative injury indicator malondialdehyde(MDA)were analyzed.(5)Inflammatory factors and apoptosis-related factors: inflammatory factors including IL-6 and TNF-? and apoptosis-related factors Bcl-2 and Bax were detected.Results:(1)The blood pressure and urinary protein of the L-NAME-induced PE-like rats were significantly higher than those of the controls.Placental weight,body weight and length,and tail length of the fetal rates were significantly reduced in the PE-like model,suggesting the PE-like model was successfully constructed.After the intervention by using zinc gluconate,all the parameters were improved,and most of them returned to normal.(2)Compared with the control group,the levels of plasma antioxidant factors CAT,GSH and SOD in PE-like model rats decreased,while the peroxidation product MDA increased.After the administration of zinc gluconate,the levels of antioxidant factors recovered and the oxidative damage was markedly reduced.(3)The levels of IL-6,and TNF-? rose and the ratios of Bcl-2/Bax decreased in the blood of the PE-like model and after the intervention of zinc gluconate,the inflammatory factors and apoptosis-related indicators reversed,approaching or even reaching the level of the controls.Conclusion: Zinc gluconate can improve blood pressure,urinary proteinuria,and pregnancy outcomes in PE model rats.The mechanism of zin gluconate in PE protective effect may be related to antagonize oxidative stress,to reduce oxidative injury and inflammatory reaction,and to inhibit mitochondrial-mediated apoptosis.
