Objective This topic studies the promotion of angiogenesis with QDTM Formula,using the model of vascular injury of transgenic zebrafish and the model of human umbilical vein endothelial cells to promote angiogenesis.It provides modern basic research theory support for clinical application of QDTM Formula in the treatment of coronary heart disease.Methods1.Induction of transgenic zebrafish(vascular endothelial cell green fluorescence marker)fish line Tg(Fli-1: EGFP)y1 using endothelial growth factor receptor tyrosine kinase inhibitor VRI to establish an internode vascular injury model in zebrafish.After the model was established,Add QDTM formula and mono-flavor decoction for drug treatment and intervention.Mono-flavor drugs are HQ,DS,CX,HZ,SZ.The experimental design group was divided into blank control group(using zebrafish embryo culture solution as blank control group),model group(VRI induction),and different concentrations(VRI + QDTM,VRI + HQ,VRI + DS,VRI +CX,VRI + HZ).,VRI + SZ),after drug intervention 24 h,the restoration effect of different concentrations of drugs on the internode blood vessels of VRI damage was observed under a fluorescence inversion microscope.2.Using zebrafish(green fluorescent marker of vascular endothelial cells)fish line Tg(Fli-1: EGFP)y1,a fully fertilized embryo of 24 h was developed and exfoliated under a microscope.Directly joined QDTM formula and mono-flavor decoction for drug intervention,mono-flavor drugs are HQ,DS,CX,HZ,SZ.After the drug intervention 48 h,the effect of different concentration of the drug on the venous growth of zebrafish was observed.3.Using HUVEC cells,the human umbilical vein endothelial cells were used to establish a cell injury model through cell scratches,and different concentrations of QDTM formula were intervened to observe the promotion of endothelial cell migration by different concentrations of drugs.4.The expression of transmigration-related proteins in endothelial cells was detected by immunoimprinting.Result1.Compared with the control group,the internode blood vessels of zebrafish in VRI group decreased significantly,indicating that VRI induced the model of internode blood vessel injury of zebrafish was established successfully.QDTM formula had a certain repair effect on the blood vessels damaged by zebrafish,and 5 mg/ml had a statistically significant difference(P<0.01).There was no significant statistical difference in the repair of VRI-induced internode vascular injury of zebrafish with HQ,DS,CX,HZ and SZ.2.Compared with the control group,QDTM formula had sprouting and crossing points increased.The effect was significant at a concentration of 5 mg/ml(p<0.001).HQ group had sprouts and increased interpoints on the subenteric venous vessels and the effect was obvious at 5 mg/ml concentration(P <0.05).DS group had sprouts and increased interpoints on the subenteric venous vessels,and the effect was significant at 5 mg/ml concentration(P <0.01).CX group had sprouts and increased interpoints on the subenteric venous vessels,and the effect was obvious at 5 mg/ml concentration(P<0.001).SZ group sprouted and increased the intersection points of the subenteric vein vessels,and the effect was obvious at 2 mg/ml concentration(P < 0.05).3.QDTM formula has a catalytic effect on HUVEC cell migration.4.QDTM formula can increase the expression related proteins P38(P < 0.01),AKT(P< 0.01)Conclusions1.QDTM formula,guided by the theory of Traditional Chinense Medicine,can be used to intervene in the treatment of coronary heart disease by promoting angiogenesis to establish blood circulation and promote endothelial cell migration.2.QDTM formula can promote the sprouting of blood vessels under the zebrafish intestine and have a recovery effect on the damage of blood vessels in zebrafish.3.QDTM formula has a catalytic effect on HUVEC cell migration.4.The mechanism of QDTM formula promote vascular growth may be related to the increase of the expression of P38 and AKT...
