Fluorescent probe is a kind of organic small molecule which will change its fluorescent properties aiming to detect dynamic level with the binding to specific molecule,ion,protein and nucleic acid.Comparing to traditional detection methods,the fluorescent probe is superior in convient operation,high sensitivity and excellent selectivity applied in fields widely,such as biochemistry,physiology and medical.As one of the important sulfhydryl amino acids in the in vivo,Cysteine plays the significant role in a variety of life activities.Abnormal levels of Cys have been associated with a lot of diseases such as cardiovascular disease,liver damage,Alzheimer's disease and Parkinson's disease.Therefore,it is obvious important for clinical diagnosis and research for biochemical mechanism to develop efficient tools to detect the concentration of Cys in cell and creature.In the second chapter,we design and synthesis a novel Cys sensor Cys W-1 to detect Cys through the cleavage reaction between the two fluorphore benzothiazole and nitrobenzoxadiazole with the fluorescent intensity enhancements in the wavelength of 455 nm and 565 nm.The probe has excellent fluorescent properties including the high sensitivity and selectivity to Cys,the low biotoxicity according to the experiment results in He La cells and L02 cells.What's more,there is a steady white light emitting system of Cys W-1 formed by the optimization to several parameters like excitation wavelength,voltage and response time.The value of the study is that the fluorescent sensor Cys W-1 can not only detect Cys in various environment,but also form white light emitting by the mixture of fluorescence under certain conditions.It is a more functional tools for the diagnosis and prevent revelant diseases and it is an attempt to structure novel complicated fluorescent system.In the third chapter,we design and synthesis a series compound named ISOPH trying to introducing ortho group to realize the bi-functional optimization of Cys fluorescent probe in fluorescence-enchancing and bathochromic-shift.The probe ISOPH-2 improves correlative fluorescent properties by this easier and more general methods.The sensor ISOPH-2 has good fluorescent properties including the fluorescent stability over 24 hours,wide detection conditions between p H from 7.0 to9.0,excellent biocompatibility,high sensitivity with the LOD of 0.072 ?M and good selectivity,In addition,the probe also successful application in cells and nematode.The strategy attempts to solve the dilemma fluorescent-enhhancing and bathochromic-shift providing reference value to fluorescent probe design and study.In summary,basd-on several fluorophores,this research complished two work on Cys probe provding reference for structure design and the basis of design strategy.Also,the study supplies the new research methods for exploiting of Cys sensors with a broad prospect on application and development.
