Rhizoma acori tatarinowii(Latin name:Acorus tatarinowii Schott),a gramineous perennial herb of the genera Araceae and Calamus,collects its dried roots in autumn and winter and is the most widely used medicinal part.The traditional Chinese medicine is rich fragrant,warm,and has the effect of dissolving dampness,dispersing knot and tonifying liver and kidney.It is found that ?-asarone and a-asarone are the main pharmacological components in the volatile oil of Acorus tatarinowii.These two active components have effects on cardiovascular system,respiratory system,digestive system,central nervous system and so on.For example:reduce blood lipids,anti-intestinal spasm,dredge spleen and stomach,calm and stop epilepsy,intellectual and brain,protect neurons and so on.In this study,the volatile oil in the rhizomes of Acorus tatarinowii from different producing areas in Anhui Province was determined by GC-MS.the results showed that the volatile oil contained 25 common chemical components,such as ?-asarone,?-asarone,?-asarone,methyl eugenol,cis-methyl isoeugenol and trans-methyl isoeugenol It was found that ?-asarone and ?-asarone accounted for more than 90%of the total volatile oil and were the main active components.The contents of ?-asarone and a-asarone in volatile oil of Acorus tatarinowii were determined by GC-MS.The results showed that under the conditions of split ratio 5:1,heating rate 5? min-1 and temperature range 97?260?,the peak shape and separation effect of the analytes were good,and the analysis time was short.Through the linear regression equation of peak area,the linear range of ?-asarone was 515?5312ng·ml-1,and a-asarone was 54?620ng·ml-1.The precision test,stability test,reproducibility test and recovery test of the two components were investigated.It was found that the method was feasible.Three different extraction methods of volatile oil from Acorus tatarinowii were compared,including steam distillation,supercritical fluid extraction and headspace injection.The extraction time,the amount of water and the soaking time were investigated by orthogonal test,and the optimum extraction process was determined as extraction time The results showed that the optimum extraction conditions were:9h,7 times of water,1h of soaking time;the extraction temperature,pressure and particle size were investigated by supercritical fluid extraction,and the best process was 10 mesh sieve,30MPa and 45?;headspace sampling method mainly investigated the temperature and equilibrium time of sample bottle,and determined that the temperature of sample bottle was 100? and the equilibrium time was 30 minters.After comparison,it is found that steam distillation is the best extraction method at present.The relative content of volatile oil in 30 batches of Acorus tatarinowii from Anhui Province was determined by GC-MS.After screening,the relative content limits of ?-asarone and ?-asarone were determined.At first,the relative content of ?-asarone in Acorus tatarinowii should be controlled within 61%?75%,and the relative content of a-asarone should be controlled within 3.7%?16.8%,and the relative content of six characteristic components should be more than 70.0%The ratio of volatile oil to ?-CD,inclusion temperature and stirring time were studied.The results showed that the best inclusion process was 1:8,stirring 1.5h and 35?.The main components and relative contents of volatile oil from Acorus tatarinowii were studied by saturated water solution method.The results showed that the main components and relative contents of volatile oil had no significant changes before and after inclusion.This study found that it is reasonable and feasible to determine the main components and content of volatile oil in Acorus tatarinowii by GC-MS.the preliminary evaluation method of volatile oil components in Acorus tatarinowii produced in Anhui Province was scientific and effective.
