Background: The incidence rate of Endometrial Cancer(EC)ranks the second in the female reproductive system malignant tumors in China.And adenocarcinoma is the most common pathological type,which is increasing worldwide.Some evidence suggests the pathogenesis of EC may be related to genetic variation.However,in further investigate it hasn't been study clearly yet.Even though the prognosis of most early endometrial cancer patients is good,if the disease spreads and differentiates poorly,the prognosis will go poor.Therefore,the discovery of novel biomarkers and therapeutic targets is particularly important for early diagnosis and improvement of survival in patients with endometrial cancer.Objective: Our study aimed to investigate the expression of kdm5 a,kdm5b and FoxO1 in endometrial adenocarcinoma,paracancerous tissue,hyperplastic and normal endometrium tissue,We suppose to explore the role and significance of these proteins in endometrial adenocarcinoma.Methods: 30 specimens of endometrial adenocarcinoma confirmed by gynecological surgery in the Third Affiliated Hospital of Nanchang University from January 2015 to June 2019 were collected,Meanwhile,15 specimens of paracancer endometrial tissue,15 specimens of simple hyperplasia endometrial tissue and 15 specimens of normal endometrial tissue were selected as the control group.To detect the expression level of KDM5 A,KDM5B and FOXO1 in each group by using Immunohistochemical method(SP).Data was analysis by SPSS 26.0 statistical software.Patients were followed up in outpatient clinic and the relationship between KDM5 A,KDM5B,FOXO1 and prognosis of patients with endometrioid adenocarcinoma was analyzed.Results:1.Among the four groups,KDM5 A and FOXO1 showed the most strong expression in normal endometrial tissue.And KDM5 B had the most strong expression in endometrioid adenocarcinoma tissue.2.Spearman correlation analysis show that KDM5 A are negatively correlated with KDM5B(r=-0.709,P < 0.001).KDM5 A are positively correlated with FOXO1(r=0.515,P=0.004).What's more,the relationship between KDM5 B and FOXO1 is negatively correlated(r=-0.442,P=0.014).3.The log rank test show that the overall survival differenceen is no statistically significant,which between KDM5 A express positive cases and express negative in dometrial adenocarcinoma tissues(P = 0.735).The overall survival rate of KDM5 B express positive cases was lower than those negative cases,and the difference was statistically significant(P = 0.001).In opposite,the overall survival rate of FOXO1 express positive cases was higher than those negative cases,but the difference was no statistically significant difference(P = 0.138).Conclusion:1.The expression level of KDM5 A and FOXO1 is low and the expression level of KDM5 B is high in endometrial adenocarcinoma,suggest that theye may be involved in the pathological process of endometrial adenocarcinoma.2.KDM5 B is associated with poor prognosis of endometrioid adenocarcinoma.3.In endometrioid adenocarcinoma,the expression level of KDM5 A and FOXO1 was positively correlated,while the expression level of FOXO1 and KDM5 B were negatively correlated.
