Objective:To investigate the expression and clinical significance of TET gene and 5hm C in chronic sinusitis.Methods:Polyp tissues of 20 patients with chronic sinusitis with polyps(CRSw NP,CW),hook process tissues of 20 patients with chronic sinusitis without polyps(CRSs NP,CS)and hook process tissues of 10 patients with Normal nasal septum deviation(N)were collected.The expression of TET and 5hm C in chronic sinusitis was detected by immunofluorescence,western-blot and Quantitative real-time PCR.Results:(1)mmunofluorescence results showed that 5hmc was expressed in CRSs NP,CRSw NP and Normal tissues.Pairings showed that the fluorescence expression of5 hmc in the CRSs NP group was significantly higher than that in the CRSw NP group and the control group(P<0.05),and there was no significant difference between the CRSw NP group and the control group(P > 0.05).(2)The results of real-time fluorescent quantitative PCR by TET showed that the m RNA expressions of TET1,TET2 and TET3 were significantly increased in the CRSs NP group compared with the Normal group and the CRSw NP group(P<0.05).There was no significant difference in the expression of TET1,TET2 and TET3 in the CRSw NP group compared with the Normal group(P>0.05).(3)The results of TET western-blot analysis showed that the expression of TET1 and TET3 proteins in the CRSw NP group was lower than that in the control group(P<0.05),while the expression of TET2 proteins was not significantly different(P >0.05).Compared with the CRSw NP group and Normal group,the expression of TET1,TET2 and TET3 proteins in the CRSs NP group increased(P<0.05).Conelusion:Compared with the control group,the expressions of 5hm C,TET1,TET2 and TET3 in the nasal mucosa of CRSs NP patients were abnormally increased,while the expressions of TET1 and TET3 proteins in the CRSw NP group were decreased,suggesting that TET mediated DNA demethylation may play a role in the occurrence of CRS.
