Study On The Antidepressant Effects Of Extracts From Akebia Trifoliata And Uncaria Sinensis In CUMS Model Mice

Objective: To study the antidepressant effect of extracts from Akebia trifoliata and Uncaria sinensis on chronic unpredictable mild stress model(CUMS)mice,explore the mechanism of antidepressant action,and compare the antidepressant effect of Akebia trifoliata and Uncaria sinensis extract.Methods: 56 male mice were randomly divided into 7 groups: blank group,model group,fluoxetine group(0.3 mg/mL),Akebia trifoliata 0.5-fold group(1.125 g/mL),Akebia trifoliata 2.0-fold group(4.5 g/mL),Uncaria sinensis 0.5-fold group(1.125 g/mL),Uncaria sinensis 2.0-fold group(4.5 g/mL);the depression model of mice was established by CUMS.After 30 days of modeling,the body mass,sugar water preference,forced swimming immobility time and hanging tail immobility time of mice were tested.The extracts of Akebia trifoliata and Uncaria sinensis were obtained by water extraction and alcohol precipitation.After the successful modeling,the fluoxetine group,Akebia trifoliata 0.5-fold group,Akebia trifoliata 2.0-fold group,Uncaria sinensis 0.5-fold group and Uncaria 2.0-fold group were given respectively mice in the 2.0-fold group were given gavage continuously for 15 days(the amount of gavage was 0.4 mL/20g),the body weight,sugar water preference,forced swimming immobility time and tail suspension immobility time of mice were measured;the contents of 5-HT,NE and DA in brain tissue of mice in each group were measured by enzyme-linked immunosorbent assay(ELISA);Western blot(Western blot)The expression of 5-HT1A receptor in hippocampus of mice in each group was detected by blot.Results: Compared with the blank group,the model group mice's body weight and sugar water preference decreased significantly,forced swimming time and tail suspension time increased significantly(P<0.05),which indicated that the model was successful;behavioral test results showed that after 15 days of administration of Akebia trifoliata and Uncaria sinensis extract,compared with the model group,the fluoxetine group,Akebia trifoliata 0.5-fold group and Akebia trifoliata 2.0-fold group Compared with the model group,fluxetine group,Akebia trifoliata 0.5-fold group,Akebia trifoliata 2.0-fold group,Uncaria 0.5-fold group and Uncaria sinensis 2.0-fold group showed significant increase in body weight and sugar water preference,forced swimming immobility time and tail suspension immobility time were significantly reduced(P<0.05),indicating that Akebia trifoliata and Uncaria sinensis can significantly improve the depression symptoms of mice;The results of ELISA showed that the contents of 5-HT,NE and DA in brain of mice in fluoxetine group,Akebia trifoliata 0.5-fold group,Akebia trifoliata 2.0-fold group,Uncaria 0.5-fold group and Uncaria 2.0-fold group were significantly higher than those in model group,indicating that Akebia trifoliata and Uncaria sinensis can improve the levels of 5-HT,NE and DA in brain and alleviate depression symptoms;Western blot results showed that compared with the model group,the expression of 5-HT1A receptor in the hippocampus of mice in fluoxetine group,Akebia trifoliata 0.5-fold group,Akebia trifoliata 2.0-fold group,Uncaria 0.5-fold group and Uncaria sinensis 2.0-fold group was significantly increased(P<0.05),indicating that Akebia trifoliata and Uncaria sinensis could upregulate the expression of 5-HT1A receptor in the hippocampus of mice,and improved the content of 5-HT in the brain,and played an antidepressant role.Conclusion: Akebia trifoliata and Uncaria sinensis could obviously improve the depression symptoms of cums model mice.The mechanism was related to the increase of monoamine neurotransmitters such as 5-HT,NE and DA in the brain of mice,and the increase of the expression of 5-HT1A receptor in the hippocampus of mice.The concentration of 4.5 g/mL of Akebia trifoliata and Uncaria sinensis could achieve the same antidepressant effect as fluoxetine.The antidepressant effect of Akebia trifoliata was slightly stronger than that of Uncaria sinensis.
