Study On The Hypoglycemic Effect And Mechanism Of Black Onion Polysaccharide In T2DM Rats

As a traditional plant resource,onion has been found to have many medicinal effects in recent years.In this paper,the hypoglycemic activity of black onion polysaccharide on rats with type 2 diabetes mellitus(T2DM)was studied,and the mechanism of its hypoglycemic action was preliminarily studied.T2DM rats were induced by alloxan(ALX).After successful induction,the rats were divided into control group,model group,intermediate hyperglycemia group and hyperglycemia group.The black onion polysaccharide solution was given to the rats in the intermediate hyperglycemia group and hyperglycemia group.Meanwhile,the rats in the control group and model group were given an equal volume of normal saline.And gavage was once a day for 4 weeks.During this gavage,blood glucose and body weight of the rats were measured regularly every week.The results showed that compared with the control group,the weight of rats in the model group was significantly lower than that in the control group,and the difference was statistically significant(P<0.01).Compared with the model group,the body weight of rats with intermediate hyperglycemia group and hyperglycemia group was significantly higher than that of the model group,and the difference was statistically significant(P<0.01).Compared with the model group,there was no significant change in body weight of rats in the hyperglycemia group(P>0.05).In addition,the fasting blood glucose of the rats in the middle and high glucose group gradually returned to normal as the gavage progressed,and the blood glucose value of the rats in the intermediate hyperglycemia group and hyperglycemia group returned to normal after only one week of gavage,which was significantly lower than the rats in the model group(P<0.01).After continuous gavage for three weeks,the blood glucose level of the rats in intermediate hyperglycemia group maintained close to the normal blood glucose level instead of continuous declination.The fasting blood glucose of rats in the hyperglycemia group also decreased gradually with the progress of gavage and decreased from 27.2 mmol/L to 14.2 mmol/L in one week after gavage with a decrease of about 47.8% in blood glucose,showing there is a significant hypoglycemic effect on the T2 DM rats.The subsequent decrease of blood glucose slowed down with intragastric administration.After the completion of intragastric administration in the fourth week,the fasting blood glucose of rats in the highglucose group was 10.8mmol/L.After four weeks of intragastric administration,fasting blood glucose decreased significantly(P<0.05),indicating that black onion polysaccharide had a good hypoglycemic effect.In this study,the effects of black onion polysaccharide on liver and kidney function,lipid metabolism and related parameters of T2 DM were studied.The results showed that the levels of Alanine aminotransferase(ALT),Aspartate aminotransferase(AST),Creatinine(CRE),Blood urine nitrogen(BUN),Total cholesterol(TC),Triglyceride(TG)and Low density lipoprotein cholesterol(LDL-C)were decreased and the level of High density lipoprotein cholesterol(HDL-C)increased in the rats of the intermediate hyperglycemia group and hyperglycemia group after gavage,indicating that there is a protective effect of black onion polysaccharide on the liver and kidney functions of T2 DM rates.In order to further clarify the protective effects of the black onion polysaccharide,the rats of executed after gavage around pathological detection of relevant organs.The results showed that the pancreas,liver and kidney of rats in the intermediate hyperglycemia group and hyperglycemia group gavages with black onion polysaccharides all improved and repaired to varying degrees.Around after the determination of lavage rats biochemical and pathological indicators testing shows that black onion polysaccharide on rat liver,kidney,pancreas has certain repair and protection function,and then using reverse transcription polymerase chain reaction and protein immunoblot method of rat pancreas,the liver and kidney tissue mRNA and protein expression,clarifying the black polysaccharide onion improve the molecular mechanisms of diabetes and related metabolic diseases.After four weeks of intragastric administration,the mRNA and protein expression levels of the apoptotic gene bax were significantly increased in rats of the model group.However,the gene and protein expression levels of the apoptotic gene bcl-2 were significantly increased in the medium and high glucose group and rats in the high glucose group after intragastric administration of black onion polysaccharide,which significantly inhibited apoptosis.The above results indicate that black onion polysaccharide can reduce blood glucose concentration in T2 DM rats,improve lipid metabolism in type 2 diabetic rats,repair and protect the liver,kidney and pancreas of diabetic rats,and thus reduce the risk of diabetic complications.
