Objective:The experimental study of Kunming mice observed aerobic exercise and blackberry LBP alone and in combination with the intervention of chronic cerebral ischemia mice and Notch channel related factors in the brain and blood were compared between the differential expression of comparative analysis.Methods:Fifty male KM mice were randomly divided into model group(M group)and model group(including SW,LBM group,LB group and swimming group LRM + SW),positive control group(BLDJ)and sham-operated group(SO).After modeling,each intervention group on the third day of moderate intensity swimming and black apocynum(200mg/kg)gavage intervention for a total of 7 weeks.Neurological behavioral assessment was performed in each group after modeling and neurobehavioral assessment was performed during the intervention.After the experiment,the mice were fasted overnight and the animals were sacrificed to collect blood and brain tissue was isolated.Elisa method was used to detect the protein level of NSE and S100B in serum.Nissl staining was used to observe the morphological changes of brain tissue.The protein levels of Notchl,Jaggedl,NCX1,SYP and CaBP-D28k in brain tissues were detected by immunohistochemistry.Quantitative technique was used to detect the expression of Notchl,Jaggedl,NCX1,SYP and CaBP-D28k in brain tissues and blood.Results:?Whole brain appearance observation:M group mice right atrium showed a large area of atrophy,and cerebral cortex blood vessels significantly reduced the extent of atrophy in the intervention group was significantly reduced,including LRM,BLDJ appear more clearly macular.Compared with M group,there was a significant difference in LRM atrophy degree in LRM + SW group(P<0.01).?Neurobehavioral evaluation showed that the scores of brain tissue injury in M group were the highest(P<0.05),the scores of SW>LRM>BLDJ>LRM + SW were lower than those in intervention group.?Elisa immunosorbent assay showed that the concentration of NSE in M group was the highest,the concentration of SWF group and LRM + SW group was the lowest(P<0.01).The concentration of S100B in M group was significantly higher than that in SW group(P<0.05).The concentration of S100B in LRM +SW group was significantly lower than that in M group(P<0.01).?The results of Nissl staining showed that the neurons in M group had a large area of damage,and the extent and extent of neuronal damage in each group were decreased to some extent.The difference was statistically significant(P<0.01).?Immunohistochemistry showed that the expression of Notchl,Jaggedl,NICD,SYP,NCXl and CaBP-D28k in M group was significantly lower than that in intervention group(P<0.01),and the protein expression of each factor in LRM + SW group was higher than that in M group Interfered with SW,LRM group(P<0.05).?Real-time PCR results showed that compared with M group,mRNA expression of Notchl,Jaggedl,NICD,SYP,NCXl and CaBP-D28k in tissues and blood increased(P<0.05,P<0.01)The expression level of Notchl,Jaggedl and NCX1 in brain tissue and blood had a moderate correlation(r>0.5),and the expression level in tissues was higher than that in blood(P<0.01)<0.05).Conclusions:Aerobic exercise and blackberry lycium barbarum polysaccharides significantly alleviate brain injury induced by chronic cerebral ischemia in mice and promote neuronal repair to some extent,and the combined effect is more significant.The effect may be through regulating Nocth pathway Related nerve repair factor expression,thereby inhibiting brain injury.As a noninvasive and early detection method,the expression level of RNA in plasma has a good application prospect for the screening of markers in the development of such diseases.
