Systems Pharmacology-based Analysis Of The Action Mechanisms Of Fuzheng Xiaoliu Decoction Against Colon Cancer

Cancer has become the number one killer that threatens human life,and colon cancer,as a common malignant tumor,has the development trend of higher incidence and younger patients,which has brought a huge burden to society and people’s lives.The onset of colon cancer is hidden and difficult to detect,and colon cancer is prone to distant metastasis.And most of them have developed to the middle or late stage at the time of diagnosis.At the time of diagnosis,most patients have progressed to the middle or late stage.At present,its pathogenesis is not clear,and it is generally believed that its etiology is related to factors such as diet,disease,and genetics.The current treatment methods mainly use methods such as surgery and chemotherapy.Modern medical treatments have some deficiencies and limitations in the treatment of colon cancer,and the efficacy of traditional Chinese medicine(TCM)formulas in cancer treatment has been confirmed.Studies have shown that in the treatment of malignant tumors of the digestive tract,supplemented with Fuzheng Xiaoliu Decoction has obvious clinical effects.The use of Fuzheng Xiaoliu Decoction can effectively control the development of the disease,improve immunity,reduce adverse reactions,and has high safety,but its exact treatment mechanism is still unclear.This study mainly used systematic pharmacological methods that combined absorption,distribution,metabolism and excretion(ADME)screening,as well as drug targeting,network,pathway analysis and other technologies to analyze the action mechanism that the Chinese medical formula Fuzheng Xiaoliu Decoction can effectively treat colon cancer from different levels.The main research contents and results of this article are as follows:(1)All the compounds in 7 herbs of Fuzheng Xiaoliu Decoction was got from TCMSP database,74 active compounds were obtained through OB and DL parameter screening,and 104 drug efficacy targets related to colon cancer diseases were obtained;(2)Target GOBP enrichment analysis found that the biological processes corresponding to these targets are mostly related to colon cancer diseases;(3)The construction of the compound-target interaction network has obtained a series of active molecules and drug efficacy targets that may play an important therapeutic role,such as Quercetin,Apigenin,Luteolin,Ursolic acid,Kaempferol and other active compounds as well as ESR1,AR,NOS2,PTGS2,GSK3 B and other efficient targets;(4)the construction of the target-disease-disease categories interaction network found that drug targets are closely related to 23 types of colon cancer-related diseases,including colonic neoplasms,neoplasm metastasis,colorectal neoplasms,inflammation,pain,gastrointestinal hemorrhage,etc.And divided these 23 diseases into 7 categories;(5)Construction of targetpathway interaction network,obtained 10 biological pathways closely related to colon cancer,including PI3K-Akt signaling pathway,HIF-1 signaling pathway and MAPK signaling Pathways,etc.(6)The integration of the "colon cancer disease pathway" has resulted in three typical treatment modules: apoptosis module,inflammation module and angiogenesis module.(7)Differential expression analysis and survival analysis of target genes showed that 39 genes were down-regulated and 65 genes were up-regulated in colon cancer.PLAU,BIRC5,FOS,ESR1,etc.have different expressions in liver cancer or lung cancer,and the active molecules in Fuzheng Xiaoliu Decoction may act on such targets and participate in the prevention and treatment of colon cancer metastasis.The low expression group of CYP19A1 showed good survival status.Comprehensive systematic pharmacology provides a clear method to illustrate the molecular mechanism of Fuzheng Xiaoliu Decoction in the treatment of colon cancer.Multiple molecules in Fuzheng Xiaoliu Decoction act on multiple targets,synergistically regulate multiple biological pathways,and play a role to achieve the therapeutic effect on colon cancer by promoting apoptosis,inhibiting inflammatory response,inhibiting angiogenesis and other ways simultaneously.System pharmacology provides indeed feasible theories and methods for the modernization of TCM.It will help to comprehensively,systematically,profoundly and thoroughly reveal the interaction mechanism between TCM and organisms from the molecular level and the system level.This will also promote the application of traditional medicine in modern medicine.
