Abstract/Summary:
Objective:Cervical cancer is one of the major cancers leading to high mortality of women worldwide,and its incidence is the first among malignant tumors of female reproductive tract.Among them,cervical squamous cell carcinoma accounts for the most,but in recent years,studies have found that cervical adenocarcinoma patients gradually increase,and the majority of young patients.Compared with cervical squamous cell carcinoma,cervical adenocarcinoma of the radiotherapy is relatively poor.Therefore,chemotherapy is particularly important in the treatment of cervical adenocarcinoma.Chemotherapy resistance is a major obstacle in the treatment of cervical adenocarcinoma,which has become one of the main factors affecting the curative effect,recurrence and survival rate of patients.In this study,the expression of KYNU protein and P-GP protein in cervical adenocarcinoma tissue and their correlation were discussed,and the relationship between the expression of KYNU protein and P-GP protein and the 5-year survival rate of patients with cervical adenocarcinoma was analyzed,so as to provide reference for further exploration of the mechanism of platinum resistance of cervical adenocarcinoma.Methods:1.Data collection:(1)Collected in Jiangxi Province maternal and child health care in January 2003-December 2008 between the first stage Ⅰ b2 ~ Ⅱ b finish two process on the basis of the platinum neoadjuvant chemotherapy and 37 patients with cervical adenocarcinoma were diagnosed by pathology and clinical follow-up data of 37 patients with cervical adenocarcinoma were reference to therapeutic effect evaluation standard of solid tumor(2000)is divided into sensitive to chemotherapy group and drug resistance,and there were 13 patients,24 cases.(2)Consulting the relevant medical records and treatments of patients,collect the basic information of patients’ age,CT imaging data before and after neoadjuvantchemotherapy,biopsy pathological data before treatment and postoperative pathological data,etc.2.Immunohistochemical(pv-9000)staining was used to detect the expression of KYNU protein and p-gp protein in the biopsy tissues of 37 cases of cervical adenocarcinoma.The number of positive cells was observed under light microscope.Immunohistochemical results grading standards for reference literature:-,+,+ +,+ ++,which defines "-,+" as lower expression,"+ +,+ + +" is defined as a high expression.3.The expressions of KYNU protein and p-gp protein in all cases were analyzed,as well as the differences in the expressions of KYNU protein and p-gp protein in the platinum-based chemotherapy drug sensitive group and drug resistance group.4.The correlation between the expression of KYNU protein and p-gp protein in cervical adenocarcinoma was analyzed,and the relationship between them and cervical adenocarcinoma was preliminarily discussed.5.Analyzing the relationship between the expression of KYNU protein and p-gp protein and the clinicopathological features of cervical adenocarcinoma and the5-year survival rate of patients with cervical adenocarcinoma.Result:1.The positive expression rates of KYNU protein and p-gp protein in cervical adenocarcinoma tissues were 86.5%(32/37)and 83.8%(31/37),respectively.2.The high expression rate of KYNU protein in the drug-resistant group was70.8%(17/24),and that in the sensitive group was 30.7%(4/13),with statistically significant differences(P < 0.05).The high expression rate of p-gp protein was 83.3%(20/24)in the drug-resistant group and 30.7%(4/13)in the sensitive group,the difference was statistically significant(P < 0.05).3.There was a positive correlation between the expression of KYNU protein and p-gp protein(r=0.386,P < 0.05).4.KYNU proteins with clinicopathological features of cervical adenocarcinoma no significant correlation(P > 0.05),P-gp protein with cervical adenocarcinoma patients age,pathological type,pathologic stage,lymph node metastasis,nosignificant correlation(P > 0.05),and the tumor is closely related to the clinical stage(P < 0.05).5.The 5-year survival rates of patients with high and low expression of KYNU protein in cervical adenocarcinoma were 61.9% and 93.8%,respectively,with a statistically significant difference(P < 0.05).The 5-year survival rates of patients with high and low expression of p-gp protein in cervical adenocarcinoma were 75.0% and84.6%,respectively,with no statistically significant difference(P > 0.05).Conclusions :1.High expression of KYNU protein may be related to drug resistance of cervical adenocarcinoma.2.High expression of p-gp protein is associated with drug resistance in cervical adenocarcinoma.3.The KYNU protein is associated with the p-gp protein.4.There was no significant correlation between KYNU protein and the clinicopathological features of cervical adenocarcinoma,while P-gp protein was associated with the clinical stage of cervical adenocarcinoma.5.The expression of KYNU protein is related to the prognosis of patients with cervical adenocarcinoma.
