ObjectiveTo Observe,under the condition of oxygen-glucose deprivation(OGD),the mRNA expression of Peroxiredoxin 1(PRDX1)and Transforming growth factor-?1(TGF-?1)that are Cytokines released by astrocytes.And investigate whether the Rhy would influence the mRNA expression of astrocytes secreted factor PRDX1 and TGF-?1 after OGD.MethodsThe hippocampal tissue of sprague-daw rats within 72 hours after birth was collected,and the primary astrocytes in the hippocampal region were extracted.After cell culture to the fourth generation,the purity of astrocytes was determined by immunofluorescence.The model of oxygen-glucose deprivation was established with anaerobic tank and sugar-free balanced salt solution.Set up four groups,including Control,OGD,rhy high-dose group(0.2 mg/ml)and rhy low dose group(0.02 mg/ml).After reoxygenation for 24 h,the total RNA of each group was extracted,and the expression of Prdx1 and TGF-?1 mRNA of astrocytes in each group was determined by fluorescence quantitative PCR.Results1.The cells extracted from the hippocampus can be filled with the bottom of the culture bottle every 7-9 days on average,and astrocytes are the main ones.2.The purity of astrocytes cultured to the fourth generation can reach more than 90%.3.The expression of PRDX1 and TGF-?1 mRNA in astrocytes was up-regulated after OGD1h/R24 h.4.The mRNA expressions of PRDX1 and TGF-?1 of astrocytes in the rhy high-dose group were down-regulated compared with the OGD group,and the mRNA expressions of PRDX1 and TGF-?1 of astrocytes in the rhy low-dose group were down-regulated compared with the OGD group.The down-regulation effect of Rhy on PRDX1 mRNA expression in astrocytes was more significant in the Rhy high-dose group than the low-dose group.Futhermore,compared with the Rhy low-dose group,the high-dose group had a more significant down-regulation effect on the expression level of TGF-?1 mRNA in astrocytes.Conclusions1.The purity of astrocytes purified by trypsin digestion was higher.2.The expression levels of PRDX1 and TGF-?1 mRNA in astrocytes increased after OGD1h/R24 h.3.Rhy can down-regulate the expression levels of PRDX1,TGF-?1 mRNA in astrocytes after OGD1h/R24 h.
