Breast cancer is the most prevalent malignant tumor among women in the world.In recent years,with the increasing importance on breast cancer prevention and the continuous development of cancer treatment methods,the number of women who die of breast cancer every year has declined compared with previous years,but the age of onset has been declining,which seriously endangers the physical and mental health of almost all age women.Therefore,further exploring the specific mechanism of the occurrence,development and metastasis of breast cancer may provide more help for the prevention,diagnosis,treatment and prognosis of breast cancer.Transcription factor FOXM1 has been found to be closely related to many tumors,especially to breast cancer.It has been found to be highly expressed and affects the phenotypic transformation of breast cancer cells: promoting the proliferation and metastasis of breast cancer cells;promoting epithelial-mesenchymal transformation of breast cancer cells;promoting the self-renewal of breast cancer stem cells and increasing the Stem cell characteristics of cancer cell;influencing the prognosis of patients.In recent years,with the further study of FOXM1,it has been found that FOXM1 is also involved in the regulation of intracellular redox signals.Redox reaction is one of the most important molecular event in biochemistry.Many studies have shown that redox signal transduction is closely related to cardiovascular diseases,tumors,diabetes,neurodegenerative diseases,aging,chronic inflammation and so on.It has become a hot research field in the world.PRDX1 is an important oxidoreductase in organisms.It plays an important role in cell redox signal transduction and tumorigenesis.In this paper,the interaction between FOXM1 and peroxidase PRDX1 was proved by mass spectrometry analysis and protein immunoprecipitation experiments.The mutants of FOXM1 were constructed,and it was found that the two proteins bind to each other through the formation of disulfide bond by 539 cysteine on FOXM1.At the same time,the high expression of PRDX1 can enhance the transcriptional activity of FOXM1,and high expression of FOXM1 can promote PRDX1 to enter the nucleus,which lays a foundation for further study of the functional changes of FOXM1 after oxidation modification and the mechanism of their interaction on the occurrence and development of breast cancer.
