Background:Breast cancer is one of the most common malignant tumors in modern women.At present,there are more than one million new cases of breast cancer every year in the world,and the incidence of young breast cancer is increasing year by year.Breast cancer has been a serious threat to the physical and mental health of women.In recent years,the development of gene sequencing and molecular biology has provided a new way for the early detection and treatment of breast cancer.In the process of biological evolution,cells continue to evolve and produce corresponding regulatory mechanisms,according to which the control of cell cycle is based on certain norms.In the process of cell cycle regulation,many genes and proteins are involved,and cell division cycle related factor 3(CDCA3)is one of them.CDCA3 is a necessary cytoplasmic protein in cell mitosis.Analyzing the expression of CDCA3 in breast cancer tissue and its effect on the function of breast cancer cells may bring new therapeutic targets for the treatment of breast cancer.Object:To study the expression of CDCA3 in breast cancer tissues and breast cancer cell lines,and to analyze the relationship between the expression of CDCA3 and clinical prognosis,cell proliferation,etc.,in order to find new prognostic factors or therapeutic targets.Methods:1.The cancer tissues and paracancerous tissues of 40 patients with breast cancer who underwent surgery in our hospital from September 2017 to January 2018 were collected.2.The expression of CDCA3 in breast cancer and paracancerous tissues was analyzed by immunohistochemical method,and the experimental results were statistically analyzed combined with clinical and pathological data.3.The expression of CDCA3 in breast cancer cell lines was analyzed byrealtimePCR.4.Celigo cell count,apoptosis detection and MTT were used to detect the effect of CDCA3 on the proliferation of breast cancer cells.Results:1.The expression of CDCA3 in breast cancer tissues was higher than that in paracancerous tissues,and the difference was statistically significant.(P < 0.05).2.The expression level of CDCA3 had no significant difference in age,tumor size,pathological tissue type and molecular type(P > 0.05),but was correlated with lymph node metastasis and TNM stage(P < 0.05).3.CDCA3 is highly expressed in breast cancer cell lines MDA-MB-231 and MCF7,after three days of shRNA lentivirus infection,the mRNA expression of CDCA3 gene in MDA-MB-231 and MCF7 cells was inhibited(P < 0.01).Through the detection of Celigo,MTT,flow cytometry apoptosis,compared with the control group,it was found that after interfering with the expression of CDCA3,the apoptosis of MDA-MB-231 and MCF7 cells in the experimental group increased and the proliferation rate was significantly inhibited(P < 0.01).Conclusion:The expression of CDCA3 is increased in breast cancer tissues and breast cancer cells,and the high expression of CDCA3 is associated with worse prognosis;CDCA3can inhibit breast cancer cell apoptosis and promote breast cancer cell proliferation;CDCA3 may become a new prognostic factor for breast cancer.
