| Objective:Qizhi Tongluo Capsule is a Chinese patent medicine for the clinical treatment of sequelae of stroke,and its curative effect is exact.In order to explore its pharmacological action components,this study uses a combination of decoction and Chinese medicine serum medicinal chemistry to treat Qizhi Tongluo Capsule.Studies on the medicinal flavors of phlegm and phlegm-relief,and anti-spasmodic and antispasmodic effects of the capsules are conducted,which lays a foundation for the basic research of the medicinal substance of the prescription.Methods:?1?SD rats were divided into the full-prescription group,Huatan Kaiqiao,Xifeng Tongluo group?Huatan Tongluo group?,Group of deficiency of Huatan Kaiqiao,Xifeng Tongluo?Deficiency of Huatan Tongluo Group?,model group,and sham operation group.A total of five groups were used to establish a model of cerebral ischemia-reperfusion injury in rats by using the suture method.The neural function score of the model rats was used to determine the area of cerebral ischemia and determine oxidative stress indicators such as superoxide dismutase?SOD?,glutathione peroxidase?GSH-Px?activity,malonaldehyde?MDA?content,and measurement of inflammatory response indicators,such as tumor necrosis factor-??TNF-??,interleukin-1??IL-1??content,observe the brain tissue HE stained sections and other indicators to reflect the pharmacodynamic effect of the chemical group;?2?using Ultra Performance Liquid Chromatography?Ultra Performance Liquid Chromatography,UPLC?to conduct basic research on medicinal substances,establish fingerprints of ten batches of medicinal materials through liquid phase conditions,identify common peaks,and establish single-drug,negative,control,blank serum,U-containing serum UPLC atlas,through the comparison of each atlas,analyze the transitional components in the medicine-flavored blood of the Huatan Tongluo group and assign its source to infer the substance basis of the medicine-flavored medicine in the Huatan Tongluo group.Results:?1?Compared with the sham operation group,the activity of SOD and GSH-PX in the model group decreased significantly,and the neurobehavioral score,cerebral infarction area,MDA,IL-1?,and TNF-?contents increased significantly?P<0.05?,indicating that the brain The ischemia-reperfusion rat model was successfully established.Compared with the model group,the deficiency prescription group,the Huatan Tongluo group and the whole prescription group can significantly reduce the neurobehavioral score,cerebral infarction area,MDA,IL-1?,TNF-?content and increase SOD in cerebral ischemia-reperfusion rats.And GSH-Px activity?P<0.05?,in which the deficient group and the Huatan Tongluo group reduced the neurobehavioral score,cerebral infarction area,MDA,IL-1?,TNF-?content of cerebral ischemia-reperfusion rats and increased SOD There was no significant difference in the effect of GSH-PX activity?P>0.05?,but the deficiency group reduced the neurobehavioral score,cerebral infarction area,MDA,IL-1?,TNF-?of cerebral ischemia-reperfusion rats compared with the whole prescription group.content,which significantly reduced the activity of SOD and GSH-Px?P<0.05?,indicating that the medicinal taste of Huatan Tongluo group played an important role in the whole prescription.?2?The final UPLC chromatographic conditions were determined as follows:Agilent Zorbax SB-C18 chromatographic column?100mm×2.1mm,1.8?m?,mobile phase methanol?A?-0.1%phosphoric acid water?B?,mobile phase gradient elution conditions?010min,5%13%A,1015min,13%A;1540min,13%40%A;4060min,40%75%A;7578min,75%78%A;7880min,78%80%A?,The injection volume is 1?L,the detection wavelength is 210nm,the column temperature is 30°C,and the flow rate is0.3mL·min-1.36 common peaks were matched.The results showed that the No.3 peak was uracil,the No.4 peak was hypoxanthine,the No.12 peak was gastrodin,the No.14 peak was p-hydroxybenzyl alcohol,four of the 36 peaks were only contributed by Gastrodia elata,five peaks were only contributed by lard soap,and the rest peaks were contributed by two or more herbs.There are 6 kinds of transitional components in the blood,among which 2 are prototypes and 4 are metabolic components.Conclusion:The taste of Huatan Tongluo Group plays an important role in the whole prescription of Qizhi Tongluo capsule,which further shows the rationality of the prescription.It is found that there are six kinds of blood migration components in Huatan Tongluo Group,which further clarifies the material basis of the effect of Qizhi Tongluo Capsule.At the same time,through this study,it shows that it is feasible to study the material basis of the efficacy of traditional Chinese medicine. |
