Objective:To explore the mechanism of how QiZhi capsule to treat vascular dementia and lay the foundation for the further research and development.Methods:1. Effect of QiZhi capsule on ET-1and CGRP in vascular dementia rats brain tissueWistar rats establish the vascular dementia rat model. The experiment set six groups:model group, western medicine group, high dose traditional Chinese medicine group, low dose Chinese traditional medicine group, shame operation group and the normal group. The rats was prepared with medicine administration by gavage according to the following plan:high dose Chinese medicine group with QiZhi capsule’s4g/kg, low dose Chinese traditional medicine group with QiZhi capsule2g/kg, western medicine group with hydergine0.3mg/kg, normal group, the model group and shame operation group are given volumetric physiological saline, once a day,30days in succession. After gavage, put the rats to death, take brain tissue and grout centrifugal, take supernate, determine ET-1 and CGRP in brain tissue using radiation immune analysis.2.Effect of QiZhi capsule on hypoxia HUECV-304activity and ICAM-1Wistar rats serum containing medicine preparation:rats random into three groups, each group of5only. Gavage administration according to the following plan:Chinese traditional medicine group with QiZhi capsule12g/Kg, positive control group with hydergine0.27mg/Kg, and the control group was given volumetric physiological saline. Each rat was drenched for five times. One hour after the last time of the gavage, collect heart blood, serum separation, inactivated processing for sparing.KM mice medicated serum preparation:mouse randomly divided into five groups, and each group of20only. Gavage administration according to the following plan:high, medium and low doses Chinese medicine groups were given QiZhi capsule18g/kg,13.5g/kg and4.5g/kg separately; Positive control group with hydergine0.4mg/kg, given the volume control such as physiological saline. Each mouse was drenched for five times. One hour after the last time of the gavage, collect orbit blood, serum separation, inactivated processing for sparing.MTT method to determine the influence of medicated serum on human umbilical vein endothelial cell activity, ELISA method to determine the change of human umbilical vein endothelial cells ICAM-1expression.3.Statistics analysisVariance analysis was accomplished using SPSS13.0statistical software, all the data was signified in mean±standard deviation (±s), P<0.05for differences with a statistical significance. Results:Compared with model group, ET-1content decreased and CGRP content increased obviously in high and low dose traditional Chinese medicine model group rats brain tissues. Medicated serum makes human umbilical vein endothelial cells in a low state surial was significantly increased, ICAM-1secretion decline. Differences are significant.Conclusions:QiZhi capsule may play role in the treatment of vascular dementia by adjusting the ET-1and CGRP balance, reducing vascular endothelial cells ICAM-1secretion, and enhancing the ability of resisting lack of oxygen of endothelial cells and so on.
