Role Of Abnormal Detrusor Cx43 Distribution And Expression In Bladder Formation Of DCP

Objective: To detect the distribution and expression of connexin43(Cx43)in detrusor tissue in guinea pig diabetic cystopathy(DCP),and to study the role of Cx43 in the formation of diabetic bladder.Methods: Sixty healthy male guinea pigs aged 2-3 months were randomly divided into a experimental group(n = 40)and a normal control(NC)group(n = 20).In the experimental group,guinea pigs were injected intraperitoneally with Streptozotocin(STZ)dissolved in p H 4.4 and concentration of 0.1 mmol / L sodium citrate to induce diabetic guinea pig models.Diabetic bladder guinea pigs were screened into the DCP group(n = 10),guinea pigs without diabetic bladder were included in the NDCP group(n = 10),and the NC group(n = 10)was given a single intraperitoneal injection of sodium citrate Solution.The pathological changes of bladder detrusor muscle were observed by HE staining in DCP group,NDCP group and NC group.The location and distribution of Cx43 in bladder detrusor muscle were observed by immunofluorescence staining.The expression of Cx43 protein in bladder detrusor tissue of the three groups was detected by western blot.Results: HE staining revealed that the morphology of the bladder detrusor muscle bundles in the guinea pigs of the DCP group was relatively irregular,and the distance between the muscle fibers was widened and sparsely distributed.The morphology of bladder detrusor muscle bundles of guinea pigs in NDCP group and NC group was regular,and the muscle fibers were arranged closely.The results of immunofluorescence staining showed that Cx43 in the detrusor tissue cell membrane and cytoplasm was specifically stained with red,indicating that Cx43 was mainly expressed in the cell membrane and cytoplasm,and no Cx43 expression was seen on the nucleus.Compared with the NDCP group and the NC group,the intensity of Cx43 fluorescent staining in the detrusor tissue of guinea pigs in the DCP group was significantly lower.The difference in the intensity of positive staining between the NDCP group and the NC group was not significant.Western blot showed that the expression of Cx43 protein(0.52 ± 0.02)in bladder tissue in DCP group was lower than that in NC group(0.68 ± 0.02)and NDCP group(0.70 ± 0.03)(P <0.01).The expression of Cx43 protein in the bladder tissue between the NDCP group and the NC group was not significantly different(P >0.01).Conclusion: Decreased expression of Cx43 in bladder detrusor may be one of the important factors leading to the onset of DCP.
