A Study On The Expression Of HnRNP K In Different Pathological Stages Of Chronic Liver Disease

Objective: The purpose of this study was to determine the expression level of nuclear heterogeneous ribonucleoprotein K in blood,urine specimens and Peripheral venous blood mononuclear cells of healthy subjects,patients with chronic hepatitis B hepatitis,post-hepatic cirrhosis,and patients with hepatitis B-related primary liver cancer,and explore the correlation between hn RNP K and PHC.Method:(1)50 cases in health subjects(HE group),43 patients with chronic hepatitis B hepatitis(CHB group),35 cases in the post-hepatic cirrhosis group(HC group)and 53 patients with hepatitis B-related primary liver cancer group(PHC group)were collected from those who attended our hospital from August 2018 to October 2019.ELISA was used to detect the expression of hn RNP K in serum and urine samples.the correlation between hn RNP K and PHC was evaluated.(2)Include 15 patients in each of the HE group,HC group,CHB group and PHC group;Quantitative Real-time PCR was used to detect the expression level of hn RNP K gene in PBMC,to study the gene expression characteristics of hn RNP K protein in PBMC.Results:(1)Serum hn RNP K is expressed in the HE,HC,CHB,and PHC groups,and the results of the HE,HC,and CHB groups are higher than those of the HE group.The CHB group has the highest value.The overall comparison was significantly different(H = 10.25,P <0.01);the ROC curve was plotted in the PHC group.At a MMY of 0.412,AUC = 0.775,sensitivity(Sen)was 0.769,and specificity(Sp)was 0.643.(2)Urine hn RNP K was expressed in the HE group,HC group,CHB group,and PHC group,and the results of the HE group,HC group,and CHB group were higher than those of the HE group.The CHB group had the highest value,and the overall comparison was significant(F = 10.58,P <0.01);ROC curve was made by measuring the hn RNP K level of each urine specimen in the PHC group.AUC = 0.652 when YI was 0.286,Sen and Sp were 1.00 and 0.286,respectively.(3)Draw a ROC curve based on the blood / urine hn RNP K ratio to evaluate its utility in diagnosing PHC.When the YI is 0.398,AUC = 0.695,Sen and Sp are 0.769 and 0.629,respectively.(4)Sen and Sp in diagnosis of PHC were increased to 90.62% when serum AFP and serum hn RNP K were parallel and sequenced;Sen and Sp were increased to 100% when combined with urine hn RNP K.(5)The relative expression of hn RNP K m RNA in PBMC samples was up-regulated in the CHB group,HC group,and PHC group and all were higher than those in the HE group,with the highest value in the CHB group(P <0.05).The overall distribution and comparison between groups,There were significant differences(F = 254.72,P <0.001).conclusion:(1)The expression of hn RNP K in the PHC group was up-regulated in serum and urine samples with better Se and Sp;combined with changes in the hn RNP K blood / urine ratio can help diagnose PHC.(2)The expression of hn RNP K in serum and urine samples combined with serum AFP can increase the Se and Sp of PHC diagnosis.(3)Different expression of hn RNP K m RNA in PBMCs of four groups of subjects,further affirming the correlation between hn RNP K and PHC...
