Objective:In this study,through measuring the contents of 1,25?OH?2D3,HIF-1?,IL-17 and?-SMA in serum of healthy subjects and patients diagnosed with NAFLD?Nonalcoholic fatty liver disease?and analyzing the changes in these factors in NAFLD patients with different fibrosis degrees,as well as the difference in the clinical indicators between the two groups,in order to investigate the changing tendency of these indicators in the development of hepatic fibrosis in order to speculate its possible mechanism to provide a new target for the prevention and treatment of fibrosis progression in NAFLD.Methods:In this study,we selected 57 patients with NAFLD as the experimental group and according to the NFS?NAFLD fibrosis score?criteria,divided them into three groups:Group A,Group B,Group C.And 16 healthy subjects in the same period were selected as control group.The serum contents of 1,25?OH?2D3HIF-1?,IL-17and?-SMA in the experimental and control group were measured by ELISA.And changing features of these indicators in different groups and difference of PLT,ALT,AST,GGP,ALB between experimental and control group were analyzed.Results:1.Compared with the experimental group,the average ages and BMIs of the control group were lower with statistically significant difference?P<0.05,P<0.01?.And there was no statistically significant difference in sex between the two groups?P>0.05?.2.Compared with the control group,the contents of ALT,AST and GGP in the experimental increased while the contents of ALB and PLT decreased,and the difference between the two groups was of statistical significance?P<0.01?.3.Compared with the control group,the contents of 1,25?OH?2D3 in the experimental group were lower while the contents of HIF-1?,IL-17 and?-SMA were higher.And there was statistically significant difference between the two groups?P<0.01?.4.The content of 1,25?OH?2D3 in the experimental group decreased while the contents of HIF-1?,IL-17 and?-SMA gradually increased with the increasing severity of hepatic fibrosis.And the difference between the two groups was of statistical significance?P<0.05?.5.1,25?OH?2D3 was negatively correlated with HIF-1?,IL-17 and?-SMA in the experimental group?r=-0.933,-0.939,-0.939,P<0.01?Conclusions:1.Compared with the control group,the serum levels of 1,25?OH?2D3in the experimental group were lower and decreased with the increasing severity of hepatic fibrosis,which is related to inflammation and hypoxia in the body,suggesting that 1,25?OH?2D3 could be used to assist in the assessment of liver fibrosis degree,and may play a protective role in the progression of liver fibrosis in NAFLD,providing new thoughts for the NAFLD therapies.2.Compared with control group,the expression levels of HIF-1?,IL-17,?-SMA in the serum of the experimental group were higher and correlated with the severity of hepatic fibrosis in NAFLD,suggesting that HIF-1?,IL-17,?-SMA can be used to predict the severity of NAFLD clinically.3.1,25?OH?2D3 was negatively correlated with HIF-1?,IL-17 and?-SMA in the experimental group,suggesting that Vitamin D may slow the progression of hepatic fibrosis by improving tissue hypoxia and reducing the body's inflammatory response.
