Objective:To preliminarily explore whether EphA2 is involved in the occurrence of PDR and analyze the correlation between EphA2,VEGF concentration and systemic metabolic indexes such as blood glucose,HbA1c and blood lipids.We hope to provide new ideas and theoretical basis for the prevention and treatment of the disease.Methods:A total of 40 patients with 40 eyes in the PDR group and the Control group were included in the ophthalmology inpatients of Hospital of Ophthalmology from September 2018 to November 2019.Among them,20 eyes of 20 patients with type 2 diabetes in the PDR group,20 eyes of 20 patients in the control group,including 13 eyes with Idiopathic Macular Epiretinal membranes,4 eyes with idiopathic macular hole,and 3 eyes with vitreomacular traction syndrome.Serum and vitreous specimens of the above patients were collected separately,all patient demographic information and basic clinical data were recorded,and the contents of EphA2 and VEGF in serum and vitreous were detected by ELISA.SPSS 18.0software Statistical analysis was performed on each group of data.Results:1.The serum VEGF and EphA2 concentrations in the control group were57.34(40.88,88.80)pg/mL and 18.11(8.65,31.62)pg/mL.The serum VEGF and EphA2 concentrations in the PDR group were 118.91(68.63,208.41)pg/mL and41.56(27.85,50.31)pg/mL.The concentrations of VEGF and EphA2 in serum in the PDR group were significantly higher than those in the control group(P<0.05).2.The concentrations of vitreous VEGF and EphA2 in the control group were 94.79(47.29,128.03)pg/mL and 18.67(11.26,39.59)pg/mL.The concentrations of EphA2 were1381.18(830.61,1752.88)pg/mL and 49.49(39.30,83.29)pg/mL.The concentrations of VEGF and EphA2 in vitreous in the PDR group were significantly higher than those in the control group(P<0.05).3.The concentration of VEGF in vitreous of PDR group was higher than that of serum VEGF(P<0.05).The concentration of EphA2 in vitreous of PDR group was higher than that of serum EphA2(P<0.05).4.There was a positive correlation between VEGF concentration and EphA2concentration in serum of the PDR group(r_s=0.865,P=0.000),There was a positive correlation between VEGF concentration and EphA2 concentration in vitreous of the PDR group(r_s=0.732,P=0.000).5.Compared with the control group,the fasting blood glucose,BUN,and Cre were significantly higher in the PDR group,while the smoking history,Drinking history and clinical indicators such as BMI,systolic blood pressure,diastolic blood pressure,ALT,AST,and UA were not statistically different.6.The VEGF concentration in the vitreous of patients in the PDR group was positively correlated with Cre of patients in the PDR group(r_s=0.707 P=0.000).Conclusion:1.The occurrence and development of PDR may be related to the high expression of VEGF and EphA2 in the serum and vitreous of PDR patients.2.There may be a correlation between high levels of EphA2 expression and high levels of VEGF expression in the serum and vitreous of PDR patients.3.Fasting blood glucose,BUN and Cre may be risk factors for PDR.The appearance of diabetic nephropathy and elevated creatinine may be indicative of aggravated diabetic retinopathy.
