Objective: To investigate the protective effects of D-limonene on lipid metabolic in ethanol-induced liver injury in rats and to study it`s prevention and treatment mechanism.Methods: 70 male Wistar rats were randomly divided into 7 groups. Group A was the control group given distilled water daily. Group B was alcohol model group. C, D, E groups were D-limonene low, medium and high dose intervention ones. Group F was water and D-limonene group. G group was Diammonium Glycyrrhizinate control group.Experiment last for 6 weeks. After the last administration, rats were fasted 12 h, chloral hydrate anesthesia, abdominal aortic blood serum and liver tissue specimens were separated. HE staining of liver tissue for morphology; electron microscopy studied the ultrastructural changes in liver tissue. To detect ALT, AST, ALP, Ch E,TG, CHO, LDL-C.Results: HE staining showed structural integrity in group A liver tissue, with cell nucleus located in the central of the cell. Group B liver tissue was bullous steatosis and a large number of inflammatory cell infiltrated. Group C bullous steatosis in visibility, as well as fat around the central vein and a large number of inflammatory cell. Group D had a lot of vacuoles,fatty degeneration formed a few large lipid droplets and a small amount of inflammatory cell infiltrated. E group the sizes of the liver cells were consistent, no significant lipid droplets, a few vacuoles; infiltration of a small amount of inflammatory cells. Group F liver cell sizes were consistent, no liver cell degeneration or necrosis morphological changes. Group G liver cell sizes were consistent, a small amount of vacuoles, no liver cell degeneration or necrosis morphological changes.Group A electron microscope showed normal liver cell structure, round or oval nucleus, mitochondria with normal morphology. Group B liver cells many secondary Lysosomes, deformed mitochondria and many lipid droplets. Group C shows a small amount of lipid droplets,Golgi normal,mitochondria clear. Group D small amount of lipid droplets, mitochondria normal. Group E mitochondria and Golgi were normal; small amount of lipid droplets,cell nucleus were round. Group F organelles were without exception; small amount of lipid droplets. Group G a small amount of lipid droplets; lysosomes can be seen occasionally; mitochondria normal.Conclusion: This research is prepared through rigorous design and operation, and the results are authentic. HE staining showed structural integrity in groups with D-limonene and the liver tissue is well protected with cell nucleus located in the central of the cell.D-limonene can protect liver tissue injured by too much alcohol. |
