Objective:Esophageal cancer is characterized by insidious onset,high degree of malignancy,poor prognosis and low survival rate,Oxidative stress is closely related to the occurrence and development of many tumors including esophageal cancer.The genes associated with the oxidative stress may become a new target for potential anti-tumor therapy.The nuclear facto-erythroid 2-related factor 2(Nrf2)is a key factor in cellular oxidative stress response.This study aimed to investigate the expression of Nrf2 in esophageal squamous cell carcinoma and its relationship with clinical stage,and to analyze the application prospect of Nrf2 inhibitors in cancer.Studies have shown that DJ-1 controls the cytoplasmic stability and final nuclear translocation of Nrf2,so this study explored the expression of DJ-1 in esophageal squamous cell carcinoma tissue and its relationship with clinical stage,and analyzed whether there is a correlation between the expression level of DJ-1 and the expression level of Nrf2.The content of trace element zinc in the human body is less than one ten thousandth,but it plays an important role in maintaining the normal physiological and biochemical processes of the body.Therefore,this study explored the relationship between the content of trace element zinc in the blood of patients with esophageal squamous cell carcinoma and its clinical stage,and explored whether Nrf2 and trace element Zn are related in the pathogenesis of esophageal squamous cell carcinoma.Methods:Collect pathological specimens of patients with esophageal squamous cell carcinoma treated in the First Hospital of Jilin University from September 2015 to August 2018.The expression of Nrf2 and DJ-1 in cancer tissues was detected by immunohistochemistry.Comparison of counting data among groups was conducted by χ2 test or Fisher’s exact test,and the results were processed by SPSS21.0 statistical software.The test level was α=0.05;At the same time,the content of trace element Zn in the blood of these patients and the content of trace element Zn in the blood of healthy people were counted.The results were processed by t-test of mean comparison.The test level was α=0.05 on both sides.When P<0.05,it was considered statistically significant.Results:1.In 46 cases of esophageal squamous cell carcinoma,the positive expression rate of Nrf2 was 10.87%.Clinical observation showed that in esophageal squamous cell carcinoma tissues with I,II,III and IV stages,the positive expression rate of Nrf2 was not statistically significant.2.In 46 cases of esophageal squamous cell carcinoma,the positive expression rate of DJ-1 was 91.30%.Clinical observation showed that in esophageal squamous cell carcinoma tissues with I,II,III and IV stages,the positive expression rate of DJ-1 was not statistically significant.3.In this study,the expressions of DJ-1 and Nrf2 in patients with esophageal squamous cell carcinoma were significantly negatively correlated.4.The Zn content in the blood of patients with esophageal squamous cell carcinoma was significantly lower than that in healthy people.The content of Zn in the blood of patients with stage IV esophageal squamous cell carcinoma was significantly lower than that of patients with stage II、stage III esophageal squamous cell carcinoma.Although the Zn content in the blood of patients with stage III esophageal squamous cell carcinoma is lower than that in patients with stage II esophageal squamous cell carcinoma,but after statistical treatment,the difference was not significant.5.Because only five of the forty six patients with esophageal squamous cell carcinoma had a positive Nrf2 expression,considering the insufficient sample size,it was impossible to determine whether there is a correlation between the level of Nrf2 expression and the lack of trace element Zn.Conclusions:1.The expression rate of Nrf2 in esophageal squamous cell carcinoma is relatively low,and it does not increase with the increase of clinical stage.2.The expression rate of DJ-1 in esophageal squamous cell carcinoma is high,and there is no significant difference in the positive expression rate of DJ-1 in esophageal squamous cell carcinoma tissues of stage I,II,III,and IV.3.The expression levels of DJ-1 and Nrf2 in patients with esophageal squamous cell carcinoma are significantly negatively correlated.4.The Zn content in the blood of patients with esophageal squamous cell carcinoma is significantly lower than that in healthy people,and the Zn content in the blood of patients with esophageal squamous cell carcinoma is lower as the clinical stage increases.5.Because only five of the forty six patients with esophageal squamous cell cancer had a positive Nrf2 expression,considering the insufficient sample size,it was impossible to determine whether there is a correlation between the level of Nrf2 expression and the lack of trace element Zn.
