ObjectiveTo investigate the expression levels of YWHAZ in lung adenocarcinoma and pleural effusion.MethodsFirst,we collected clinical information related to tissue chips,patients with lung adenocarcinoma,and patients included in BPE or MPE.Second,we collected 35 samples of cancer tissue,adjacent tissue,and normal tissue,acquired on the basis of the inclusion and exclusion criteria.Then we collected 16 cases of BPE and MPE for the purpose of making ROC,which gave a prediction of the values of YWHAZ in BPE and MPE.Furthermore,we observed the expression levels of YWHAZ using RT-qPCR and IHC in order to analyze the relationship between YWHAZ and clinical characteristics,and then detected YWHAZ in the tissue chip using IHC.Third,a Cox regression was done to determine whether YWHAZ was an independent factor affecting prognosis in lung adenocarcinoma.Results(1)There was no difference between BPE and MPE in terms of gender,age,and smoking history.(2)The RT-qPCR results for lung adenocarcinoma showed that the YWHAZ mRNA in the lung-adenocarcinoma tissue was higher than that in matched adjacent tissue and normal tissue(P<0.001).A Spearman correlation analysis of the YWHAZ showed that it was positively correlated with CYFRA-211(P<0.05),and the Kaplan-Meier survival curve showed that the 1-year survival rate of patients in the YWHAZ low-expression group was 94.4%,higher than that of those in the YWHAZ highexpression group.IHC revealed that YWHAZ was mainly expressed in the nucleus and cytoplasm.Furthermore,the expression level of YWHAZ in lung-adenocarcinoma tissue was higher than that in either paired adjacent tissue or normal tissue(P<0.001).YWHAZ had higher expression in a variety of kinds of cancer tissue than in the paired adjacent tissue among the tissue chips,while the highest expression of YWHAZ was in lung adenocarcinoma.The expression level of YWHAZ mRNA detected using RT-qPCR showed that the YWHAZ mRNA in MPE was higher than that in BPE(P<0.05).There was a statistically significant difference when YWHAZ was used for distinguishing BPE as opposed to MPE,with a sensitivity of 75% and specificity of 93.7%.The area under the ROC curve was 0.904,as revealed using ROC(P<0.05).(3)A Cox regression analysis of YWHAZ mRNA revealed that YWHAZ mRNA was an independent factor in the survival rates of patients with lung adenocarcinoma.The YWHAZ mRNA high-expression group was at a 3.498 times higher risk of death than the YWHAZ mRNA low-expression group of lung-adenocarcinoma patients.Conclusion(1)In stage-3 lung adenocarcinoma,the expression level of YWHAZ is higher than before,and the 1-year survival rate is lower.YWHAZ mRNA is an independent factor affecting the prognosis in lung adenocarcinoma.YWHAZ may play a role as an important oncogene in lung adenocarcinoma,which would make it relevant to the prognosis.(2)YWHAZ seems promising as a molecular marker distinguishing BPE from MPE. |
