A Study On MicroRNA-451a/YWHAZ Regulating Proliferation, Migration, Invasion Of The Lung Adenocarcinoma A549 Cells

Objective: The aim of this study was to explore microRNA-451a/YWHAZ affected the biological function of lung adenocarcinoma A549 cells.Methods:(1)We used lentivirus overexpressing microRNA-451 a and its negative control to infect A549 cells and after stable generations,used puromycin for drug screening to obtain stable cell lines.Then,qRT-PCR detected lentivirus infection staining rate.Meanwhile,we studied the impact of overexpressing microRNA-451 a in cell proliferation,colony formation and invasion potential;(2)Next,we prodicted the protential target of microRNA-451 a was YWHAZ through Targetscan and miRDB;(3)Finally,We studied the influence of silencing YWHAZ in cell proliferation,colony formation and invasion potential.Results:(1)after overexpression of microRNA-451 a in A549,CCK8 detected cell proliferation was inhibited(P < 0.05);The results of cell colony formation assay showed that the number colony formation in microRNA-451 a group(100.30±10.03/plate)was lower than in oe-NC group(174.21±10.30/plate)(P<0.05);The results of wounding healing assay showed that the healing rate in microRNA-451 a group(34.19±2.13%)was significantly inhibited than in oe-NC group(51.06±4.23%)(P < 0.05);The results of cell migration and invasion assays showed that the number of cell migration in microRNA-451 a group(20.70±1.59/field)was lower than in oe-NC group(79.45±6.90/field)(P<0.05),the number of cell invasion in microRNA-451 a group(50.47±3.90/field)decreased than in oe-NC group(98.50±7.31/field)(P<0.05);(2)Targetscan and miRDB predicted that microRNA-451 a and YWHAZ had binding sites;in A549 cell lines overexpressing microRNA-45 a,YWHAZ mRNA expression level significantly decreased.(3)After silencing YWHAZ in A549,CCK8 detected cell proliferation was inhibited(P < 0.05);The results of cell colony formation assay showed that the number colony formation of si-YWHAZ group(35.11±2.35/plate)was lower than in si-NC group(56.67±4.75/plate)(P<0.05);The results of wounding healing assay showed that the healing rate of si-YWHAZ group(20.43±1.28%)was significantly inhibited than in si-NC group(30.08±2.26%)(P < 0.05);The results of cell migration and invasion assays showd that the number of cell migration in si-YWAHZ group(48.33±3.60/field)was lower than in si-NC group(77.70±5.23/field)(P<0.05),the number of cell invasion in si-YWHAZ group(50.20±3.52/field)decreased than in oeNC group(110.00±9.81/field)(P<0.05).Conclusion:microRNA-451 a might target YWHAZ to inhibited proliferation,migration,invasion of lung adenocarcinoma A549 cells.
