Background:Oral squamous cell carcinoma(OSCC)is the most common malignant tumor in oral and maxillofacial region in China,accounting for more than80%.OSCC is more common in 40-60 years old,male than female,the predilection site is seen in tongue,cheek,gum,palate,maxillary sinus and other places.OSCC often metastases to regional lymph nodes,and also to distant organs in advanced stage.There are many studies on the causes and mechanisms of OSCC,including long-term physical and chemical stimulation,viral infection,genetic factors,proto-oncogene and anti-oncogene mutations.The human body can produce an anticancer immune response through immune surveillance function,which is mainly cellular immunity.Tumor microenvironment(TME)refers to the internal and external environment that promotes the occurrence,growth and metastasis of tumor cells,which consists of interstitial cells and extracellular matrix(ECM).In the interstitial cells around TME,in addition to CD8+T cells that play an anticancer role,there are also a large number of immune Regulatory T cells(tregs).These cells can induce immune tolerance,inhibit CD8 T cells and other immune cells,secrete a variety of soluble factors to regulate the tumor microenvironment and play an important role in promoting tumor development and metastasis.As the most specific marker molecule of tregs,forkhead/winged helix transcription factor(Foxp3)plays an important role in the formation and function of tregs.Many studies have found that DCN which exists in ECM is related to tumorigenesis and development of tumor,such as liver cancer,renal cell carcinoma,colorectal cancer and so on,and some studies have shown that DCN can be used in the treatment of some tumors.DCN is a member of the leucine-rich proteoglycan family.It is directly or indirectly involved in the growth,metastasis and angiogenesis of various tumors by interacting with signal molecules.However,studies on the role of DCN and FOXP3 in OSCC have not been reported yet.Therefore,this study aims to increase more understanding of the occurrence mechanism of OSCC and explore their expression and significance in OSCC.Objective:The expression of DCN and FOXP3 in OSCC was detected to explore their significance in the occurrence and development of OSCC and their correlation,so as to add a new understanding of the pathogenesis of OSCC and find a potential treatment.Method:A total of 36 specimens from patients with OSCC were collected.All cases were diagnosed as OSCC by pathological examination.The control group consisted of 15 cases.All specimens were confirmed as normal non-inflammatory tissues by pathology.The expression of DCN and FOXP3 was detected by two-step immunohistochemical method in all cases.The expression of DCN and FOXP3 in OSCC and normal mucosa was semi-quantitatively analyzed by Imagepro-plus6.0 software.The data were statistically analyzed by SPSS22.0 software.The difference was statistically significant(p < 0.05).Result:The relative expression of Decorin in normal oral mucosa and OSCC were 0.2165±0.0797 and 0.1030±0.0739 respectively,while the relative expression of FOXP3 was 0.0631 ± 0.1365 and 0.3444 ± 0.2427 respectively.Compared with the normal control group,Decorin and FOXP3 expressions were statistically significant(P<0.05).Clinical stage III-IV and I-II comparison and differentiated degree,the relative expression of DCN had statistical significance,but there was no significant difference in lymph node metastasis;The expression of FOXP3 was significantly different in different differentiation degree and lymph node metastasis,but there was no significant difference in clinical staging.Decorin has no significant correlation with the level of FOXP3expression(r=0.303,P>0.05).Conclusion:DCN and FOXP3 are abnormally expressed in OSCC.They are related to biological processes such as invasion and metastasis of OSCC and participate in the progress of oral squamous cell carcinomas.However,no negative regulatory relationship was found between DCN and FOXP3 in this study,indicating that FOXP3 expression has a complex regulatory network,and the joint effect of the two in oral squamous cell carcinoma needs more studies.
