| Smilax China L.is a plant of Liliaceaes,also known as Jingangteng.Its medicinal parts is the dried rhizome.It received in the 2015 edition of "Chinese pharmacopoeia",with the effect of dampness turbidity,heat-clearing and detoxifying,softening and resolving hard lumps to treat urinary turbidity,excessive vaginal discharge,rheumatic arthralgia furuncle and carbuncler etc.At present,Smilax China L is mainly used for the treatment of pelvic inflammation,annex inflammation,mass and other gynecological diseases in clinically.Project previous studies showed that Smilax China L.has effective parts of anti-inflammatory and twelve components have anti-inflammatory activity.Because of Smilax China L.having many ingredients,which ingredients have anti-inflammatory activity is unclear.On the basis of previous research,this paper using the idea of spectral efficiency relationship to research the enthanol extracts of Smilax China L.in different areas,carry out chemical fingerprint,biological activity and identification of active substances based on spectral efficiency relationship for improving the quality evaluation standard of Smilax China L.and providing the basis for explaining the material basis of Smilax China L.in anti-inflammatory activity.This paper mainly completed the following research work:1 Studies on the fingerprint of Smilax China L.Collecting Smilax China L.in ten different areas,the fingerprint of Smilax China L.was established by UPLD-DAD method.The results show that there is a big difference between the 10 batch of ethanol extracts of Smilax China’s L in chemical composition.Selecting forty-five pesks from DAD fingerprints,these peaks have obvious difference in peak area,so that the different areas of Smilax China’s L.can be used in spectral efficiency study.2 Anti-inflammatory activity in vivo Using Xylene induced mouse ear swelling model to observe inhibition effect of extracts of Smilax China’s L.in acute inflammationthe with ear swelling and swelling rate.The results show that nine samples can reduce the ear swelling rate significantly compared with the model group except S4,and their anti-inflammatory activity has significant differences.Animal model of chronic pelvic inflammatory disease(CPID)was established by mechanical injury and Escherichia coli to observe the treatment of extracts of Smilax China’s L.in CPID rats by measuring SOD,MDA,IL-6and TNF-αin serum.3 Anti-inflammatory activity in vitro RAW264.7 cells was induced by LPS to observe the anti-inflammatory activity in vitro of extracts of Smilax China’s L.Investigating the consistent of experimental results in vivo and vitro to provide reference of spectral efficiency relationship.The results show that seven groups were consistent in the experimental results.It is possible to use the in vitro experiments to replace the in vivo experiments on studying spectral efficiency relationship,but more comparative experiments are needed to verify the results.4 Analysis of fingerprint with anti-inflammatory results Based on the fingerprint and in vivo pharmacological data,analyze thecorrelation between chemical composition represented by chromatographic peaks and anti-inflammatory effect by grey correlation analysis method.Analysing the correlation between forty-five peaks and six pharmacodynamic indexes and total efficacy,the results showed that correlation degree above 0.6,indicating that these chemical constituents of Smilax China’s L.and anti-inflammatory effects were associated.The strongest anti-inflammatory activity was 3# peak which the structure was unknown,chlorogenic acid,taxifolin-7-O-glucoside also has very strong correlation with anti-inflammatory activity,indicating the efficacy of traditional Chinese medicine is the synergistic effect of various chemical components. |
