Objectives:To study the inhibitory effect of the extract of Smilax trinervula Miq.(St.)on the proliferation of different tumor cells and the growth of S180 tumor-bearing mice,and to screen the anti-tumor active fractions of St..Methods:1.Alcohol extract and water extract of St.were separated by solvent method.Different parts of St.extract were extracted by different solvents.MTT method was used to investigate the inhibitory effects of different extract sites of St.on the proliferation of human hepatocellular carcinoma cell line HepG2,human lung cancer cell line A549,human breast cancer cell line MCF-7 and MDA-MB-231,human cervical cancer cell line HeLa and human ovarian cancer cell line HO-8910.2.The chemical constituents of petroleum ether fraction of alcohol extract of St.were separated by silica gel chromatography,and the inhibitory effects of each component on the proliferation of the six cancer cell lines were investigated by MTT method.3.Establish an animal model of S180 tumor-bearing mice and determine the inhibitory effect of alcohol extract of St.and its ethyl acetate fraction on the growth of S180 tumor-bearing mice.4.The expression levels of apoptosis related factors,such as caspase 3,Bax and p53,were detected by PCR.Results:1.The extracts of St.have certain inhibitory effects on different cancer cells,showing a dose-effect relationship.The semi-inhibitory concentration(IC50)of petroleum ether fraction and ethyl acetate fraction of St.alcohol extract on each cancer cell is 20-250μg·mL-1.Ethyl acetate had the strongest inhibitory effect on cancer cells.IC50 of MCF-7was 24.259μg·mL-1,that of MDA-MB-231 was 70.562μg·mL-1,and HeLa 83.577μg·mL-1.2.Component C of petroleum ether fraction of alcohol extract of St.has strong inhibitory effect on six kinds of cancer cell lines,among which the inhibitory effect on HeLa cells is the most significant,with IC50 value of 66.912μg·mL-1.3.The extract of St can inhibit the growth of S180 tumor-bearing mice,and the dosage is positively correlated with the inhibition effect.4.the expression of Bax and p53 mRNA in St.extract(0.1 g·kg-1)increased significantly(P<0.05);Conclusions:This experiment analyzed that the extract of St.has certain anti-tumor effect.The anti-tumor active components of St.may be concentrated in ethyl acetate fraction and petroleum ether fraction C.and its anti-tumor effect may be related to the up regulation of Bax and p53 mRNA expression. |
