Aims:To explore the anti-tumor activity of Periplaneta fuliginosa aqueous extract and its possible mechanism of action, it will provide a scientific basis for the future clinical application. We used in vivo and in vitro experiments to observe effects of Periplaneta fuliginosa aqueous extract in H22 tumor-bearing mice,human breast cancer cell line MCF-7 and hepatocellular carcinoma cell line Hep G2 as well.Methods:In vitro antitumor experiment: MTT experiment was used to observe the effects of different concentrations of Periplaneta fuliginosa aqueous extracts on the proliferation of MCF-7 and Hep G2 cell.In vivo antitumor experiment:To investigate the antitumor effects of Periplaneta fuliginosa aqueous extract in vivo.We constructed the H22 cell ascites tumor model, and the H22 cells were inoculated subcutaneously in Kunming mice.The mice were random Ly divided into normal group, model group,cyclophosphamide group,Periplaneta fuliginosa aqueous extract high dosage group,medium dosage group and low dosage group.Tumor growth and the organ index of the mice were observed.Pathology changes of tumor tissue was observed by HE stain. The expression of Bax and Bcl-2 protein were detected by immunohistochemical.Results:In vitro antitumor experiment: after 24 h treated with Periplaneta fuliginosa aqueous extract from each dose group compared with the control group, MTT results showed that as drug concentration increased, the rate of tumor cell growth inhibition of MCF-7 showed an upward trend, in a dose dependent manner(p<0.01). After the differert dose of the Periplaneta fuliginosa aqueous extract treated in Hep G2 cells for24 h, 48 h and 72 h, compared with the control group, All dose group can inhibit the growth of tumor cells(p<0.01)and followed with the time growth, the inhibition rate increased gradually.In vivo antitumor experiment:Compared with the model group, the tumor volume and tumor mass of the high dose group of the Periplaneta fuliginosa aqueous extract had significant difference(p<0.05).The tumor inhibition rates of the low dose group,medium dose group and high dose group of the Periplaneta fuliginosa aqueous extract, were respectively 20.72%, 29.24%, 43.16%. Compared with the normal group,the spleen index of each dose group were increased, furthmore the high and middle dose group had significant difference(p<0.05). The thymus index of each dose group was decreased, but there was no significant compared with model group(p>0.05). HEstaining showed tumor tissue have different degrees of necrosis compared with model group in the high dose group, middle dose group and low dose group of Periplaneta fuliginosa aqueous extract. Immunohistochemistry results showed that the expression of Bax increased and Bcl-2 expression decreased.Conclusion:Periplaneta fuliginosa aqueous extract has anti-tumor activity in vitro and in vivo.The anti-tumor mechanism may be related to the up-regulated expression of Bax protein and down-regulated of Bcl-2 protein expression.
