Objective:Toexplore the correlation between ApoE gene polymorphism and cognitive dysfunction in Tibetan population in Qinghai.Methods:Taking Tibetan residents in Qinghai Provincial People’s Physical Examination Hospital Center and Maduo People’s Physical as a sample,which includes 20 patients with Alzheimer’s disease,36 patients with mild cognitive impairment,and 40 healthy controls.All subjects were subject to relevant inclusion and exclusion criteria.DNA from peripheral blood of all subjects was extracted and ApoE gene polymorphism was determined by real-time quantitative PCR.Results: 1.The ApoE allele frequency and genotype of AD group,MCI group and normal control group were in accordance with H-W genetic balance law.2.The ApoE genotypes in AD group,MCI group and control group were the most common in E3 / E3 type,followed by E3 / E4 and E2 / E3 types,and no E2 / E2,E2 / E4,E4 / E4 genes were detected.In the AD group,the E3 / E3 genotype was 70.00%,followed by E3 / E4(30.00%).In the MCI group,the proportions are E3 / E3(80.56%),E3 / E4(16.66%),and E2 / E3(2.78%),and the results for the control group are the propotions are E3 / E3 genotypes(80.00%),E3 / E4(15.00%),and E2 / E3(5.00%).There was no significant difference in genotypes among the three groups.3.Among the three groups of allele frequency distribution,the ApoEε4 gene frequency in the AD group was 15.00%(6/40),which was not statistically different from the normal control group of 7.50%(6/80)(χ2 = 0,P = 1);The frequency of ε4 gene in MCI group was 8.45%(6/71),and there was no significant difference compared with the normal control group(χ2 = 0.251,P = 0.616);AD group and MCI group have no significant difference in ApoEε4 gene frequency(χ2 = 1.138,P = 0.286).Comparison of ApoEε2 and ε3 allele frequencies in the three groups showed no significant difference(P> 0.05).4.The relationship between ApoE allele and disease can be estimated by calculating the OR value.Compared with the normal control group,the ApoEε4 allele has no significant association with AD(OR = 2.177,P = 0.211),and the ε2 allele(OR = 1.513,P = 0.5518),ε3 allele(OR = 0.63,P = 0.547)were also not associated with AD.The ApoEε4 allele was not significantly associated with MCI(OR = 1.139,P = 1.0000),also,the ε2 allele(OR = 0.557,P = 1.0000),and the ε3 allele(OR = 0.1.016,P = 1.0000)have no association with MCI.The ε2 allele(OR = 1.571,P = 0.64),ε3 allele(OR = 0.62,P = 0.54),ε4 allele(OR = 1.192,P = 0.345)in AD group and MCI group)indicate that there were no significant differences in the three alleles in the AD group and the MCI group.Conclusion: There is no correlation between the apolipoprotein E(ApoE)gene polymorphism and cognitive dysfunction in Tibetans in Qinghai. |
