Objective To investigate the expression of mitochondrial autophagy protein PINK1 /Parkin in renal clear cell carcinoma and adjacent tissues,and explore its clinical significance.Methods Patients who were diagnosed with renal malignancies before surgery and received surgical treatment in patients undergoing urology in the General Hospital of Ningxia Medical University from October 2018 to October 2019 were selected.They were paired with normal tumor tissues(2cm)adjacent to cancerous tissues and fasting venous blood And related clinical data,50 cases of renal clear cells were diagnosed by pathology after operation.The expression of PINK1 and Parkin protein in cancer tissue and adjacent tissue was detected by immunohistochemical(IHC)method and(Western Blot)method respectively,and the content of ROS and MDA in serum was detected by ELISA method.Combined with the relevant clinical data,the clinical significance of PINK1 and Parkin protein in the occurrence and development of renal clear cell carcinoma was analyzed.Results The results of immunohistochemical experiments showed that the positive expression of mitochondrial autophagy protein PINK1 was brownish-yellow granules.It was mainly expressed in the cytoplasm and part of the nucleus in cancer tissues.It was mainly expressed in the proximal tubules and distal tubules of cytoplasm and adjacent tubules.In the lumen,the nucleus was not significantly expressed,and the expression was reduced compared with that in cancerous tissues.The positive expression of mitochondrial autophagy protein Parkin was brownish-yellow granules,and it was mainly expressed in the renaltubular cytoplasm and part of the nucleus in the normal renal tissue adjacent to the cancer.Cancer tissues are mainly expressed in the interstitial and part of the nucleus,and the expression is reduced compared with normal tissues adjacent to the cancer;Western-Blot results show that the expression of PINK1 in cancer tissues is significantly higher than that in normal renal tissues adjacent to the cancer(P<0.01),The expression of Parkin in normal renal tissues adjacent to cancer was higher than that in cancerous tissues(P<0.05);analysis of clinical data with patients found that the expression of PINK1 protein was not statistically significant in patients with gender,age,tumor stage,and presence or absence of lymph node metastasis(P> 0.05),and it has statistical significance with the pathological grade of tumors(P<0.05);Gender,age,tumor stage,and presence or absence of lymph node metastasis were not statistically significant(P>0.05),and were statistically significant(P<0.05)with the pathological grade of the tumor;the levels of ROS and MDA in the serum of patients before surgery increased,and the difference was statistically significant(P <0.05).Conclusion 1.The expression of mitochondrial autophagy protein PINK1 in renal clear cell carcinoma tissues is higher than that in normal adjacent tissues.The mitochondrial autophagy protein Parkin is higher in normal tissues adjacent to renal clear cell carcinomas than in cancer tissues,and acts as a tumor suppressor gene.2.Mitochondrial autophagy proteins PINK1 and Parkin are involved in the development of renal clear cell carcinoma,which may affect tumorigenesis through oxidative stress caused by ROS and MDA.
