| Purpose:In this study,SAMP in Alzheimer’s disease(AD)model mice were used as the research object to observe and study the effect of " mutual assistance of kidney and brain " electroacupuncture therapy on the expression of PINK1 and PARKIN mitochondrial autophagy-related proteins and their A proteins.To explore the specific mechanism of " mutual assistance of kidney and brain " electroacupuncture therapy on brain protection in AD model mice by reducing A protein deposition by mitochondrial autophagy.It provides a theoretical basis for clinical treatment of AD from the kidney to the brain.Material and method:18 male SAMP8 mice and 6 SAMR1 mice were chosen for the experiment.Using random number notation,6 SAMP8 mice were randomly classified into groups,Model group,Western medicine group and Electric needle groups.Also 6 SAMR1 mice were categorized as a Contrast group,Memantine and 20 mg/kg of dose were applied with Gavage,Once per day to the Western medicine group.Stimulated the electric needle group with a charged needle(frequency 2 Hz,voltage 2 V,current 0.6 MA)by acupuncturing on NephrolysIs and Taixi,for 15 minutes,once per day.The contrast group and the model group are fed normally,without any treatment.Subsequently to the eight weeks of feeding and treatment the water maze experiment was carried out on each group.After completion of behavioral tests,materials were acquired by using the Western Blot indicator to measure the self-absorption rate of mitochondria of PINK1,PARKIN.Results: 1.Water maze experiment revealed that the incubation period and swimming distance rating of the western medicine group and electric needle group were better than the model group(P<0.05),and there was no statistical difference from the model group and the contrast group(P>0.05).2.Western Block results show that the PINK1 and PARKIN in both the western medicine group and the electric needle group are lower than the model group(P<0.05),the results of the model group and the contrast group are similar(P>0.05).3.As a result of Nissl staining,the western medicine group and the electric needle group had slightly aligned nerve source cells compared to the model group,and also contained Nissl corpuscle and uniform staining.4.Immunity grouping of PINK1 and PARKIN showed distinct self-destruction in the model group,but relatively less self-destruction was observed in the control group and two treatment groups.Conclusion: 1.Electrical needles can slow down the development of memory disorders in AD model mice.2.The electric needle can be found to promote mitochondria self-destruction by enhancing PINK1 into the cell by improving mitochondria somatic potential change.3.The electric needle can cause the effects of protecting the nerve source in AD model mice based the theory and vaporization of ’mutual assistance of kidney and brain’. |
