Objective The aim of the study was to investigate effects of the suppression of inflammation and modulation of gut microbiota regulatory activity of Lycium barbarum polysaccharide(LBP)in mice with allergic asthma.Methods C57BL/6 female mice were randomly divided into four groups(15 mice /group): normal control group(Control),allergic asthma model group(OVA),asthma disease treated with LBP group(OVA + LBP)and normal control treated with LBP group(Con +LBP).The mice were treated with LBP for 2 weeks before modeling.LBP continued to interfere with mice until the end of modeling.(1)The general condition of mice(hair,lips,claws,teeth,respiratory rate,etc.)was observed.(2)He,Masson and PAS staining were used to detect the infiltration of inflammatory cells,collagen deposition and goblet cells.(3)Detection of TNF-?,MCP-1,IL-6,IL-17 A,IL-4 and IFN-? in plasma and BALF using cytometric bean array(CBA).(4)The compositions of gut microbiota in diverse groups were analyzed by high throughput sequencing of 16 S rDNA.(5)Pearson correlation analysis was used to analyze the relationship between inflammatory cytokines in plasma and gut microbiota.Results(1)Compared to OVA group,Allergic asthma symptoms in OVA+LBP group was significantly relieved,the degree of shortness of breath was lighter,and there was no bleak mouth,lips,claws and nails.(2)The pathological sections of lung in asthmatic mice showed that the infiltration of inflammatory cells,the deposition of collagen around bronchi and the proliferation of mucinous goblet cells were significantly reduced after LBP intervention.(3)CBA results showed that LBP could significantly reduce the levels of plasmaTNF-?,MCP-1,IL-6,IL-17 A and IL-4,but increased the level of plasma IFN-?.(4)Bioinformatics analysis of high-throughput sequencing showed that LBP significantly reduced the phylum levels of Firmicutes and Actinobacteria in OVA group.LBP significantly increased the genus levels of Lactobacillus,Bifidobacterium and Clostridia,and Alistipes was significantly reduced.(5)Correlation analysis showed that Lactobacillus and Bifidobacterium were positively correlated with inflammatory factors(TNF-?,MCP-1 and IL-6)respectively,and negatively correlated with IFN-?.Alistipes was positively correlated with IFN-?,and negatively correlated with TNF-?,MCP-1 and IL-6.Clostridia was not significantly correlated with inflammatory cytokines.Conclusion LBP can improve allergic asthma in mice via suppressing inflammation and regulating the composition of gut microbiota.
