Objective:To establish the model of epilepsy in rats by chemical ignition of pentaerytetrazol injection and observe the effect of liver regulation and detoxification method on seizures and emotional behavior of epileptic rats.The immunohistochemical method was used to detect and analyze the effect of liver regulation and detoxification method on the expression of NPY and its receptor Y2 in hippocampus and amygdala on epileptic rats in order to explore the neurochemical mechanism of liver regulation and detoxification method in the treatment of epilepsy,and provide an effective experimental basis for the effect of liver regulation and detoxification method in the treatment of epilepsy.Methods:60 male SD rats were randomly assigned to 10 rats as the normal group after one week of habitition,and the other rats were intraperitoneally injected with PTZ every two days,and total for 14 times.The rats of epilepsy model ware randomly assigned to the epilepsy group,the sodium valproate group,and the low dose Huazhuojiedushugan group,the medium dose Huazhuojiedushugan group,and the high dose Huazhuojiedushugan group,with 10 rats in each group.The rats in each group were kept in a single cage,and the epileptic seizures were observed and recorded during the modeling period.After the completion of the modeling,each group and the valproate group were treated with the corresponding drugs for 28 days,the control and the epilepsy groups were treated with isopyknic normal saline.After the treatment,the open field test and the elevated plus maze experiments were used for behavioral analysis.After the end of the behavioral test,immunohistochemical staining was performed to statistically analyze the positive expression of NPY and its receptor NPY2 R in the hippocampus and amygdala of the brain.Results:1.The seizures.The seizure latency in the high-dose group was significantly longer than that in the epilepsy group(P < 0.01),and the seizure grade was significantly lower than that in the epilepsy group(P < 0.01).Compared with the sodium valproate group,there was no statistical difference in the seizure latency and the seizure grade in the high-dose Huazhuojiedushugan group(P > 0.05).2.Behavioral tests.In the open field test,the duration in center,central distance traveled and entries in center of the epilepsy group were significantly less than those of the control group(P < 0.01).The duration in center,central distance traveled and entries in center of high dose Huazhuojiedushugan group were higher than those in the epilepsy group,and there was significant difference between the high dose Huazhuojiedushugan group and the epilepsy group(P < 0.01),while there was no statistical difference between the high dose Huazhuojiedushugan group and the valproate group(P > 0.05).In elevated plus maze,the entries in open arms,the duration in open arms and close arms latency of epilepsy group were less than those of control group(P < 0.01),the entries in open arms,the duration in open arms and close arms latency of the high dose Huazhuojiedushugan group were increased than those of epilepsy group(P < 0.01).There was no statistical difference between the high dose Huazhuojiedushugan group and the valproate group(P > 0.05).3.Immunohistochemistry.The expression of NPY in the hippocampal CA1,CA3,DG and central amygdala of the epilepsy group was higher than that of the control group,and there was a significant statistical difference in the CA1 area(P < 0.05),and a very significant statistical difference in other regions(P < 0.01).The expressions of hippocampal CA1 and central nucleus of amygdala in the high dose group were higher than those in the epilepsy group(P < 0.01),and the expressions of hippocampal CA3 and DG were lower than those in the epilepsy group(P < 0.05).The expression of NPY2 R in hippocampal CA1,CA3 and DG was higher in the epilepsy group than in the blank group(P < 0.01).Compared with the epilepsy group,the expression of CA1 and DG in the high dose group was higher(P < 0.05),and the expression of DG in the CA3 area was significant higher(P < 0.01).Conclusion:1.In this study,we successfully establish the epilepsy model by intraperitoneal injection of PTZ for every two days,and this seizure model has emotional disorder.2.liver regulation and detoxification method can alleviate the epileptic seizures in rats,and improve the behavior tests of open field and elevated plus maze in epileptic rats.3.liver regulation and detoxification method can change the expression of NPY and NPY2 R in the brain tissues of epileptic rats.
