Objective:To investigate the effect of elevated blood glucose on the deposition of triglycerides in the liver of rats.Methods:40 male SD rats were randomly divided into normal group(group A,n = 10),sham group(group B,n = 10),experimental group(group C,n = 10),and experimental plus intervention group(group D,N = 10).All rats were fed under the same conditions.No treatment was performed in group A.Only the spleen was removed after laparotomy in group B.Major pancreatic resection was performed after laparotomy in groups C and D,and rats in group D were treated with insulin(insulin glargine injection,0.1U / kg).,Once a day).One week and four weeks later,the four groups of rats underwent oral glucose tolerance test(OGTT).The body weight of rats was measured weekly.Blood samples were collected to measure alanine aminotransferase(ALT)and aspartate aminotransferase(AST)values.Colorimetric method was used to measure ?-glutamyl transpeptadase(GGT)value.Glyceryl phosphate-peroxide(GPO-PAP)enzymatic method to determine triglyceride(TG)value,iodine-starch colorimetric method to determine ?-amylase(?-AMS)value.Rats were sacrificed at the end of 4 weeks,liver tissues were completely removed,and tissue weights were compared while leaving GPO-PAP enzyme method was used to determine the TG content in liver tissues,and the morphology and lipid deposition of liver tissues were observed pathologically.Results:1.During the weekend of experiment 1,there was no significant change in fasting blood glucose in each group(p> 0.05).The 0.5h blood glucose in group C increased significantly(P <0.05),and the blood glucose in group C and D increased significantly at1 h and 2h(P <0.05).The area under the glucose curve(AUCG)in group C was significantly increased(P <0.05),while the increase in AUCG in group D was not significant(p> 0.05).In the weekend of experiment 4,no significant change in fasting blood glucose was observed in each group(p> 0.05).The 0.5h blood glucose in group C was significantly increased(P <0.05),the 0.5h blood glucose in group D was significantly decreased(P <0.05),and the 1h blood glucose in group C was significantly increased(P <0.05),the 2h blood glucose in group C and D increased significantly(P<0.05),and AUCG in group C and D was significantly increased(P <0.05).2.There was no significant difference in serum ALT,AST,GGT,?-AMS concentrations in the four groups of rats(P> 0.05).The liver TG content in group C was significantly higher than that in group B(P <0.05).There was a significant decrease in group C(P <0.05),and there was no significant difference in liver TG content between groups B and D(P> 0.05).3.There was no significant difference in liver weight,liver weight / body weight between groups(p> 0.05).Pancreatic weight,pancreatic weight / body weight in group C were significantly lower than those in group B(p <0.05),there was no significant difference in pancreatic weight,pancreatic weight / body weight between groups C and D(p> 0.05),and pancreatic weight between groups A and B.There was no significant difference in pancreatic weight / body weight(p> 0.05).Conclusion:After 4 weeks of elevated blood glucose caused by partial resection of the pancreas,triglyceride deposition in the liver tissue increased significantly in rats,and liver triglyceride decreased significantly after insulin intervention.
