Objective:To detect the copy number of plasma free mitochondrial DNA(mtDNA)in patients with acute pancreatitis,and to explore the changes of plasma free mtDNA copy number in patients with acute pancreatitis.To compare the relationship between plasma free mtDNA copy number and commonly used clinical indicators in patients with acute pancreatitis,and its application value in disease diagnosis,prognosis and disease assessment.Methods:Plasma samples from forty healthy subjects as control group,forty patients with acute pancreatitis(twenty patients with mild pancreatitis and twenty patients with moderate severe pancreatitis)as experimental group and six patients with acute pancreatitis on the first,third and seventh days of hospitalization were collected,and their plasma free mtDNA copy number were measured by real-time fluorescence quantitative PCR.Simultaneously detect the blood routine and related biochemical indicators of all plasma samples,compare the plasma free mtDNA copy number of the experimental group and the control group,compare the general clinical indicators of the experimental group and the control group,and analyze the correlation between the plasma free mtDNA copy number and common clinical indicators and clinical score.The changes of plasma free mtDNA copy number in patients with acute pancreatitis after hospitalization were dynamically monitored.Results:1.The number of plasma free mtDNA copies was 5.0×10~4(3.1×10~4)copies/mL in the healthy control group,1.2×10~5(1.6×10~5)copies/mL in the MAP group,7.8×10~5(5.0×10~5)copies/mL in the M-SAP group,The differences between the three groups were statistically significant(p<0.05).2.(1)The WBC of the healthy control group,MAP group and M-SAP group were4.88±0.92,12.89±3.07 and 16.54±2.39(10~9/L),respectively.The differences between the three groups were statistically significant(p<0.05).(2)The CRP of the healthy control group,MAP group and M-SAP group were2.29±2.01,22.32±16.36 and 103.80±73.95(mg/L),respectively.The differences between the three groups were statistically significant(p<0.05).(3)The PCT of the healthy control group,MAP group and M-SAP group were0.02±0.01,0.06(0.24)and 3.33(15.34)[ng/L],respectively.The differences between the three groups were statistically significant(p<0.05).(4)The TG of healthy group,MAP group,and M-SAP group were 1.47±0.16,2.46±1.46,and 7.43(28.82)[mmol/L],respectively.The differences between the three groups were statistically significant(p<0.05).(5)The HDL of the healthy control group,MAP group and M-SAP group were1.41±0.25,1.09±0.53 and 0.88±0.40(mmol/L),respectively.The healthy control group was higher than the MAP group and M-SAP group,and there was no statistical difference between the MAP group and M-SAP group.(p>0.05).(6)The LDL of the healthy control group,MAP group and M-SAP group were2.63±0.67,1.52±0.67,and 2.07±1.29(mmol/L),respectively.There was no significant difference between the three groups.(p>0.05).(7)The AMY of healthy control group,MAP group,and M-SAP group were44.83±14.72,730.44±569.63,and 715.19±550.13(U/L),respectively.Healthy control group was lower than MAP group and M-SAP group,and there was no statistical difference between the MAP group and M-SAP group.(p>0.05).(8)The LPS of the healthy control group,MAP group,and M-SAP group were23.83±6.55,1422.69±978.50,and 1216.86±738.38(U/L),respectively.The healthy control group was lower than the MAP group and M-SAP group,and there was no statistical difference between the MAP group and M-SAP group.(p>0.05).3.The copy number of plasma free mtDNA was positively correlated with WBC,CRP,PCT,TG,Ranson scores and BISAP scores in patients with acute pancreatitis.(r=0.453?0.539?0.562?0.513?0.653?0.869 p<0.05)4.The condition of patients with acute pancreatitis improved after hospitalization,and the copy number of plasma free mtDNA increased first and then decreased.Conclusion:The level of plasma free mtDNA copy number in patients with acute pancreatitis is increased,the detection of plasma free mtDNA copy number can be used in the diagnosis,disease monitoring and prognosis evaluation of acute pancreatitis.
