Study On Mitochondrial DNA Copy Number Of Peripheral Leukocyte In Patients With Alveolar Echinococcosis And Its Significance

Object:To quantify the mitochondrial DNA copy number of peripheral blood leukocytes and determine the content of cytokines and clinical index in patients with alveolar echinococcosis,and analyze the difference between case and control group with the correlation between the mitochondrial DNA copy number of peripheral blood leukocytes and inflammation indicators.Methods:36 patients with alveolar hydatidosis(case group)and 28 healthy persons(control group)were enrolled in the Department of Hepatobiliary and Pancreatic Surgery,Affiliated Hospital of Qinghai University.The peripheral blood were collected in the morning.Mitochondrial DNA copy number and cytokine content were determined by Q-PCR and ELISA,and the remaining indicators were derived from the clinical laboratory.The differences between the two groups were compared,and the correlation between the number of mitochondrial DNA copy in peripheral blood and cytokines and clinical index was analyzed.Results:1.A total of 28 persons were included in the control group,including 12 males,71.42%of Tibetans,age(46.00±12.43);36 cases,14 males,88.88%of Tibetans,age(40.72±12.31).gender(χ~2=0.103,P=0.748),nationality(χ~2=3.152,P=0.076),age(t=-1.694,P=0.095)were not different between groups.2.The number of mitochondrial DNA copy of peripheral blood leukocytes in the control group(590.57±278.81)were significantly higher than the case group(394.64±128.23)(t=-3.446,P=0.001).3.The levels of IL-2,IL-10,TNF-α,and IFN-γin the control group were(4925.32±775.55)pg/ml、(404.40±175.05)pg/ml、(53.75±5.95)pg/ml、(529.96±147.53)pg/ml,the levels of IL-2,IL-10,TNF-α,and IFN-γin the case group were(3338.64±833.68)pg/ml、(552.94±79.87)pg/ml、(39.33±7.80)pg/ml、(432.20±86.82)pg/ml.There were significant differences in each index between the two groups(P<0.05).The levels of IL-2,TNF-αand IFN-γin the control group were higher than those in the case group,while IL-10 was lower than that in the case group.4.There was no difference in ALT,AST,TBIL,DBIL,IBIL,neutrophil count and lymphocyte count between the two groups(P>0.05).Leukocyte count in case group(6.85±2.31)was higher than that in control group(5.63±1.48),P<0.05.5.Mitochondrial DNA copy number was not correlated with cytokine content and clinical indicators.Conclusion:The energy supply of peripheral leukocyte in patients with alveolar echinococcosis may be insufficient,and no correlation between each index and the number of mitochondrial DNA copies may be related to the complex immune status.
