With the development of the economy and the influence of unhealthy lifestyles,the incidence of diabetes is increasing year by year,posing a serious threat to human health.Diabetes-induced metabolic disorders and cardiovascular complications are also responsible for the high mortality rate of the disease in the world.The traditional treatment is mainly through oral hypoglycemic drugs and insulin injection.Two methods will burden the digestive system and kidneys of the human body for a long time and bring side effects.Therefore,it is very necessary to develop natural and non-toxic anti-diabetic drugs.Corn beard and dandelion,as traditional Chinese herbal medicines in China,have been wasted seriously.In recent years,due to their unique biological activities and nutrients,they have received more and more attention from the medical community and the food community,and have great development and utilization value.The experiment proves that the single corn silk extract is mainly for lowering blood sugar,and the dandelion extract has better anti-inflammatory properties.This topic is a rough extraction of corn silk dandelion,and the active ingredients of the mixed extract and its mechanism of action The preliminary research provides a theoretical basis for the direction that the mixed extract of corn beard and dandelion can resist diabetes and its complications.The total flavonoid yield is an evaluation index.Corn whisker and dandelion are mixed in a ratio of 1:2.The extraction conditions are subjected to a single factor test and orthogonal optimization.By using 70%ethanol,the ratio of material to liquid is 1:15.At60?,extract twice,2.5 h each time,to prepare a mixed extract of corn dandelion.The vitro antioxidant capacity of the mixed extract was determined by measuring the removal rate of the mixed extract for DPPH,hydroxyl and superoxide radicals.This experiment proves that the scavenging rates of the mixed extracts for these three free radicals are 68.39%,71.68%and 74.01%,respectively,and the scavenging effect is better.Among them,O2-·scavenging is the strongest and has good antioxidant capacity.Induced the construction of type ? diabetes model,and compared with the blank,negative and positive control groups to determine the effect of the mixed extract on blood glucose and biochemical indicators of diabetic rats.First,regularly observe the daily life status?body mass,mental status,etc.?of diabetic model rats.The results show that the mixed extract can improve the prominent symptoms of"three more and one less"in rats,and the high-dose group can reach the treatment level of the positive drug group?P<0.05?.Second,the measurement of rat serum found that the content of Hb and WBC increased significantly?P<0.05?,indicating that the mixed extract can effectively control the changes of blood glucose and enhance insulin sensitivity during treatment of diabetic rats.Third,the high-dose group mixed extract can significantly improve the glucose tolerance of diabetic rats and reduce the fasting blood glucose level?P<0.05?.Fourth,the medium-dose mixed extract can significantly reduce the content of TC,TG and NEFA in rats?P<0.05?,and improve the disorder of glucose and lipid metabolism in the liver of diabetic rats.Fifth,after treatment with mixed extracts of different doses by gavage,the wet weight of spleen and thymus and organ index of diabetic rats have increased,and the body's immunity has been enhanced.Sixth,the experiment found that after gavage treatment,the expression levels of immunoglobulin?IgG,IgM,IgA?in rats were somewhat higher than those in the negative model group,indicating that the mixed extract can improve the disease resistance of rats.Seventh,the mixed extract can promote the expression of anti-inflammatory factors?TNF-?,IL-1?,IL-6,IL-12?in diabetic rats,and it can promote the proinflammatory factors?IFN-?,IL-2,IL-10?Inhibition of expression can reduce the body's inflammatory response.Finally,the experiment found that the mixed extract of each dose group had no effect on the activity of AKP and LDH enzymes in rats.
